AMTS update for STTR

Advanced nuclear reactor systems currently in development include designs that utilize higher energy neutron spectrums, higher temperatures, and corrosive environments that are quite different from the current fleet of nuclear power plants in use today. As a result, new structural materials must be developed and qualified for use to support these advanced systems. For example, research is currently being conducted on a new class of materials known as multi-principle-element-alloys. These alloys show some promise to be able to meet the structural needs of these challenging reactor systems. However, due to the vast available compositional space, and the limited amount of research conducted thus far, Idaho National Laboratory (INL)sought a way to advanced mechanical testing capability for highly irradiated structural materials to increase both throughput and higher fidelity data analysis through incorporation of digital image correlation. The Automated Mechanical Testing System accomplishes these tasks by automating the testing and incorporating digital image correlation to provide higher fidelity data. INL has now received this system and is currently performing functional testing and training. INL is also performing some modifications to the system to allow for installation into a remote shielded enclosure where testing of irradiated structural materials can be performed. After testing, training, and modifications of the system are complete, INL will install the system into a shielded enclosure and begin using the system to test irradiated structural materials.

ASC CSSE Level 2 Milestone #6362: Resilient Asynchronous Many Task Programming Model

This report is an outcome of the ASC CSSE Level 2 Milestone 6362: Analysis of Re- silient Asynchronous Many-Task (AMT) Programming Model. It comprises a summary and in-depth analysis of resilience schemes adapted to the AMT programming model. Herein, performance trade-offs of a resilient-AMT prograrnming model are assessed through two ap- proaches: (1) an analytical model realized by discrete event simulations and (2) empirical evaluation of benchmark programs representing regular and irregular workloads of explicit partial differential equation solvers. As part of this effort, an AMT execution simulator and a prototype resilient-AMT programming framework have been developed. The former permits us to hypothesize the performance behavior of a resilient-AMT model, and has undergone a verification and validation (V&V) process. The latter allows empirical evaluation of the perfor- mance of resilience schemes under emulated program failures and enabled the aforementioned V&V process. The outcome indicates that (1) resilience techniques implemented within an AMT framework allow efficient and scalable recovery under frequent failures, that (2) the abstraction of task and data instances in the AMT programming model enables readily us- able Application Program Interfaces (APIs) for resilience, and that (3) this abstraction enables predicting the performance of resilient-AMT applications with a simple simulation infrastruc- ture. This outcome will provide guidance for the design of the AMT programming model and runtime systems, user-level resilience support, and application development for ASC's next generation platforms (NGPs).

Survey of Modeling and Simulation Techniques for Advanced Manufacturing Technologies Volume I – Predicting Initial Microstructures

This report summarizes the current state of modeling and simulation methods for predicting the initial structure and properties of material in components assembled using advanced manufacturing technologies (AMTs). The report is the first volume in a two-volume series. This first volume focuses on predicting initial microstructures of AM material. The second volume discusses predicting material properties given the initial microstructure. The focus is on technologies of particular relevance to the design and manufacture of nuclear reactor components. The purpose of the report is to help develop the technical knowledge base to support regulatory decisions that will be needed to assess nuclear components manufactured with AMTs as these components are installed at nuclear power plants (NPPs). The report develops a list of AMTs of particular interest to the NRC and summarizes the key microstructural features and corresponding processing parameters relevant to each technology. Further sections describe available physically-based and data-driven prediction methods and survey widely available software tools. The report concludes with a summary of gaps that may be of particular interest to the NRC when evaluating modeling and simulation methods for AMTs.

Quantitative Dissection of Agrobacterium Virulence to Generate a Synthetic Ti Plasmid

Agrobacterium is not only a costly plant pathogen but is also an essential tool for plant transformation. Though Agrobacterium-mediated transformation (AMT) has been heavily studied, its polygenic nature and complex transcriptional regulation make identification of the genetic basis of transformational efficiency difficult through traditional genetic and bioinformatic approaches. Here, we use a bottom-up synthetic approach to systematically engineer the tumor-inducing plasmid (pTi), wherein the majority of virulence machinery is encoded. Using a validated toolkit to control Agrobacterium gene expression in planta, we perform a quantitative dissection of AMT to investigate the contributions of critical vir-genes at different expression levels. We construct a synthetic pTi capable of transient plant and stable fungal transformation and characterize bottlenecks and solutions for complex polygenic synthetic pTi designs. Our reductionist approach demonstrates how bottom-up engineering can be used to dissect and elucidate the genetic underpinnings of complex biological traits, laying the foundation for future engineering to establish full synthetic control over the critical process of AMT.
