ESSENCE- A rapid, shear-enhanced, flow-through, capacitive electrochemical platform for rapid detection of biomolecules

The rapid, sensitive, and selective detection of target analytes using electrochemical sensors are challenging. ESSENCE, a new Electrochemical Sensor that uses a Shear-Enhanced, flowthrough Nanoporous Capacitive Electrode, overcomes current electrochemical sensors' response limitations, selectivity, and sensitivity limitations. ESSENCE is a microfluidic channel packed with transducer material sandwiched by a top and bottom microelectrode. The room-temperature instrument less integration process allows the switch of the transducer materials to make up the porous electrode without modifying the electrode architecture or device protocol. ESSENCE can be used to detect both biomolecules and small molecules by simply changing the packed transducer material. Electron microscopy results confirm the high porosity. In conjunction with the non-planar interdigitated electrode, the packed transducer material results in a flow-through porous electrode. Electron microscopy results confirm the high porosity. The enhanced shear forces and increased convective fluxes disrupt the electric double layer's (EDL) diffusive process in ESSENCE. This disruption migrates the EDL to high MHz frequency allowing the capture signal to be measured at around 100 kHz, significantly improving device timing (rapid detection) with a low signal-to-noise ratio. The device’s unique architecture allows us multiple configuration modes for measuring the impedance signal. This allows us to use highly conductive materials like carbon nanotubes. We show that by combining single-walled carbon nanotubes as transducer material with appropriate capture probes, NP-µIDE has high selectivity and sensitivity for DNA (fM sensitivity, selective against non-target DNA), breast cancer biomarker proteins (p53, pg/L sensitivity, selective against non-target HER2).

New generic evolution for k -essence dark energy with w ≈ - 1

We reexamine k-essence dark energy models with a scalar field Φ and a factorized Lagrangian, L = V (φ)F(X), with X = $1\over{2}$∇ µ Φ∇ µ Φ. A value of the equation of state parameter, w, near -1 requires either X ≈ 0 or dF/dX ≈ 0. Previous work showed that thawing models with X ≈ 0 evolve along a set of unique trajectories for w(a), while those with dF/dX ≈ 0 can result in a variety of different forms for w(a). We show that if dV /dΦ is small and (1/V )(dV /dΦ) is roughly constant, then the latter models also converge toward a single unique set of behaviors for w(a), different from those with X ≈ 0. We derive the functional form for w(a) in this case, determine the conditions on V (Φ) for which it applies, and present observational constraints on this new class of models. We note that k-essence models with dF/dX ≈ 0 correspond to a dark energy sound speed c$2\atop{s}$ ≈ 0.

The Essence of Reactivity

Reactive programming, functional reactive programming, event-based programming, stream programming, and temporal logic all share an underlying commonality: values can vary over time. These languages differ in multiple ways, including the nature of time itself (e.g., continuous or discrete, dense or sparse, implicit or explicit), on how much of the past and future can be referenced, on the kinds of values that can be represented, as well as the mechanisms used to evaluate expressions or formulas. This paper presents a series of abstractions that capture the essence of different forms of time variance. By separating the aspects that differentiate each family of formalisms, we can better express the commonalities and differences between them. We demonstrate our work with a prototype in Haskell that allows us to write programs in terms of a generic interface that can be later instantiated to different abstractions depending on the desired target.

Data-Driven Insights into the Structural Essence of Plasticity in High-Entropy Alloys

The heterogeneous mechanical response of a crystalline alloy with multiple principal elements was investigated using molecular dynamics simulations. The local configuration of the alloy in its quiescent state was characterized by the variables derived from the gyration tensor and the atomic electronegativity. A multivariate analysis identified the geometric and chemical factors that influenced the atomic packing variations. Further, upon straining, the non-affine displacement exhibited spatial heterogeneity. A statistical correlation was established between the local yield events and the specific features of the local configuration. Our findings, validated by the performance metrics analysis, provided a structural criterion for the instability mechanisms in high-entropy alloys (HEAs) and enhanced the understanding of their plasticity.

The Essence of Cryptol: A Denotational Cryptol Interpreter in Coq for Foundational Assurances for Quantum Resistant Cryptosystems

Systems of the utmost consequence need a means to establish authenticity of software and data. Cryptosystems implement authentication, but can be vulnerable to cryptographic and implementation attacks. With the threat of quantum cryptographic attacks, “post-quantum” cryptosystems (PQCs) must be henceforth used in these systems. However, the new cryptography needs new ways to, rigorously and machine-checkably, prove systems free of vulnerabilities. We propose a retargetable capability to rapidly instantiate proven correct postquantum cryptosystems through novel proof-carrying synthesis and proof-automation technique, extending those proven successful on existing systems. This capability is crucial to meeting the cryptographic requirements for future high-consequence systems. Since specifications for high consequence cryptography are presently captured in a domain specific language known as Cryptol. While this can enable convenient fully automated reasoning about Cryptol specificaitons and implementations via the Software Analysis Workbench (SAW), Cryptol has expressivity gaps, so that cryptosystems with probabilistic programming features like Falcon cannot be fully expressed in the language. Moreover, SAW’s automation fails for programs and specificaitons with inductive and recursive structure, as in the Sphincs+ PQC. Finally, Cryptol and SAW together represent some 200,000 lines of unverified Haskell, so that the any guarantees about high consequence cryptography are presently contingent on a large, unverified, yet trusted computing base. The first step of the larger project of agile, assured crpytography is therefore to provide a formal, mechanized semantics for Cryptol, so that the specifications expressed by cryptographers in Cryptol can be reasoned about and compiled into performant implementations with a foundational, machine checkable certificate of correctness. This report describes our work on this first step, culminating in the design of a certified denotational interpreter, in Coq, for core Cryptol.

Emergence of Relations and the Essence of Learning: A Review of Sidman's Equivalence Relations and Behavior: A Research Story

Sidman addresses two very important questions in Equivalence Relations and Behavior: A Research Story: What are the bases of behavioral competence? And how do units of learning become related? The book recounts the story of how an understanding of emergent relations and competencies was achieved through studies in his teaching-research program with mentally retarded subjects. Although children normally accrue vast networks of relations between stimuli and events, those with mental retardation typically do not. Consequently, by learning how to establish those networks, Sidman and his students contribute richly both to the cultivation of competencies by their subjects and, more generally, to an understanding of real-world human behavior. The basic equivalence paradigm affords the subject feedback and reinforcement for very specific choices during training, but the test is not for those choices! Rather, tests for equivalence look for new choices, ones seemingly quite foreign to the training regimen. The tests for equivalence relations entail presentations of stimuli that were the options for conditional choice during reinforced training. In tests of equivalence, correct choices are novel; hence, they have never been reinforced during training. The study of equivalence relations can encourage the emergence of new perspectives that are more symbiotic than competitive. In full acknowledgment of the important role and contributions made by those who identify themselves as experimental analysts of behavior, it is timely that rapprochements be worked toward, as indeed they are, to meld that perspective with others of our time. Both our research methods and our expectations about the nature of the learning process and the abilities of our subjects can delimit what they might learn and what we, in turn, learn about their learning. The text will be of great value for instruction at the upper-division and graduate levels. Its impact will be substantial, for it defines an important advance in our efforts to understand the richness of behavior in both humans and nonhuman animals. Although not presented to that end, the book might also serve to bridge communications with other groups of animal researchers whose interests lie more in a comparative or ethological framework.

Emergence of relations and the essence of learning: a review of Sidman's Equivalence relations and behavior: a research story. Book review

The author reviews and comments on the book Equivalence relations and behavior: a research story by Murray Sidman. Sidman's book reports his research about equivalence relations and competencies in children with mental retardation and how it relates to behavior. Sidman used the idea of stimulus-stimulus relations among features of the environment to develop his theories about equivalence relations. Experimental work with children and animals demonstrated their ability to use equivalence relations to learn new tasks. The subject received feedback and reinforcement for specific choices made during training, then was presented with new choices during testing. Results of the tests indicate that subjects were able to establish relations and retrieve them in different situations.

Influence of lure choice and survey duration on scent stations for carnivore surveys

Abstract Noninvasive, camera‐based, scent station surveys have become a common approach to estimate population parameters for mammalian carnivores. Many carnivores are particularly sensitive to olfactory cues; and, as such, the quality and quantity of odor attractant used during surveys can significantly affect the accuracy and precision of the survey. Additionally, survey length is also an important consideration because many mammalian predators are highly mobile and elusive in nature. Using scent station surveys in autumn 2013 and 2014 on the Savannah River Site, Aiken, South Carolina, USA, we tested the effects of 5 lures: fatty acid, fish oil, synthetic fermented egg, beaver castor, and skunk essence on visitation rates, visitation probability, and latency to visitation of multiple predator species. We also estimated occupancy probability for 2 carnivore species using different survey lengths to determine the effect of survey period on the precision of occupancy estimates. We found that skunk essence had greater visitation probability and visits than other lures evaluated. Species‐specific bait‐preference models revealed that raccoons ( Procyon lotor ) were more likely to visit skunk essence, and coyotes ( Canis latrans ) were less likely to visit fish oil as compared with fatty acid. Latency to visitation ranged from 104.7 to 112.1 hours and did not vary among scents. Longer survey durations resulted in more precise estimates of occupancy and detection probability. Our results suggest a wide range of scents commonly used for carnivore capture may perform as well as, or better than, fatty acid for noninvasive carnivore surveys. Further, study designs should carefully consider the duration of scent‐station surveys depending on the level of precision desired in estimates of occupancy and detection probability. © 2019 The Wildlife Society.

The holographic entropy cone from marginal independence

The holographic entropy cone characterizes the relations between entanglement entropies for a spatial partitioning of the boundary spacetime of a holographic CFT in any state describing a classical bulk geometry. We argue that the holographic entropy cone, for an arbitrary number of parties, can be reconstructed from more fundamental data determined solely by subadditivity of quantum entropy. We formulate certain conjectures about graph models of holographic entanglement, for which we provide strong evidence, and rigorously prove that they all imply that such a reconstruction is possible. Our conjectures (except only for the weakest) further imply that the necessary data is remarkably simple. In essence, all one needs to know to reconstruct the holographic entropy cone, is a certain subset of the extreme rays of this simpler “subadditivity cone”, namely those which can be realized in holography. This recasting of the bewildering entanglement structure of geometric states into primal building blocks paves the way to distilling the essence of holography for the emergence of a classical bulk spacetime.

Multi-level Monte Carlo methods in chemical applications with Lennard-Jones potentials and other landscapes with isolated singularities

We describe and compare outcomes of various Multi-Level Monte Carlo (MLMC) method variants, motivated by the potential of improved computational efficiency over rejection based Monte Carlo, which scales poorly with problem dimension. With an eye toward its application to computational chemical physics, we test MLMC's ability to sample trajectories on two problems — a familiar double-well potential, with known stationary distributions, and a Lennard-Jones solid potential (a Galton Board). By sampling Brownian motion trajectories, we are able to compute expectations of observable averages. These multi-basin potential energy problems capture the essence of the challenges with using MLMC, namely, maintaining correspondence of sample paths as time-resolution is varied. Addressing this challenge properly can lead to MLMC significantly outperforming standard Monte Carlo path sampling. We describe the essence of this problem and suggest strategies that circumvent diverging multilevel sample paths for an important class of problems. In the tests we also compare the computational cost of several, “adaptive,” variants of MLMC. Our results demonstrate that MLMC overcomes the collision, time scale limitation of the more familiar Brownian path MC samplers, and our implementation provides tunable error thresholds, making MLMC a promising candidate for application to larger and more complex molecular systems.

Book Review: What Makes a Good Experiment? Reasons and Roles in Science

Classification is an arduous and sometimes controversial undertaking because of its inherent cultural particularity. One famous literary example is the fictitious nomenclature of animals found in Jorge Luis Borges’s ‘The Analytical Language of John Wilkins’. According to Borges, that taxonomy was discovered by a translator, F. Kuhn, in a Chinese source and divides all animals into fourteen categories, starting with ‘those that belong to the Emperor’ and ending with ‘those that from a long way off look like flies’. Physics of the twentieth century is a ramified full-fledged technical culture, and it should surprise no one that physics experiments can be classified in different ways, allowing certain experiments to belong to more than one category. Allan Franklin’s analysis of experiments is based on his classification, which substantially grasps the essence of the experimental enterprise. Although Franklin does not offer an explicit answer to the question posed in the title, he does include a detailed examination of several important physics and biology experiments in history
