Sensitivity of puff dynamics and airborne droplet nuclei distribution to variations in violent expiration events

Here, we use large eddy simulations to investigate the puff and droplet dynamics from violent expiratory events such as coughs and sneezes in the first few seconds following an ejection. For each of the eleven simulations considered, over 60,000 droplets are ejected and individually tracked using the point-particle Euler–Lagrange approach. We test the sensitivity of the puff and droplet dynamics to various parameters including the ejection volume, momentum, and orientation. We also explore the effect of the mouth shape on the aforementioned dynamics by considering elliptical and circular inlet cross-sections. The results from the simulations compare favorably with a recent theoretical framework put forth by Balachandar et al. in terms of the puff size and propagation velocity. More importantly however, the theory is able to accurately predict the number and size spectra of the potentially virus-laden droplet nuclei that remain airborne within the puff. We observe that the ejection angle and mouth shape do not significantly affect the puff and droplet dynamics. Additionally, we quantify the carrying capacity of the detached puff portions in terms of the number and size spectra of droplets/droplet nuclei suspended within.

Using advanced data structures to enable responsive security monitoring

Write-optimized data structures (WODS), offer the potential to keep up with cyberstream event rates and give sub-second query response for key items like IP addresses. These data structures organize logs as the events are observed. To work in a real-world environment and not fill up the disk, WODS must efficiently expire older events. As the basis for our research into organizing security monitoring data, we implemented a tool, called Diventi, to index IP addresses in connection logs using RocksDB (a write-optimized LSM tree). In this work, we extended Diventi to automatically expire data as part of the data structures’ normal operations. We guarantee that Diventi always tracks the N most recent events and tracks no more than N + k events for a parameter k < N, while ensuring the index is opportunistically pruned. To test Diventi at scale in a controlled environment, we used anonymized traces of IP communications collected at SuperComputing 2019. We synthetically extended the 2.4 billion connection events to 100 billion events. We tested Diventi vs. Elasticsearch, a common log indexing tool. In our test environment, Elasticsearch saw an ingestion rate of at best 37,000 events/s while Diventi sustained ingestion rates greater than 171,000 events/s. Our query response times were as much as 100 times faster, typically answering queries in under 80 ms. Furthermore, we saw no noticeable degradation in Diventi from expiration. We have deployed Diventi for many months where it has performed well and supported new security analysis capabilities.

Basin-Wide Approaches to Hydropower Licensing

This presentation summarizes findings from a 2020 NREL technical report including the FERC relicensing process for non-federal hydropower projects, analyzes FERC-licensed projects with license expiration dates from 2018-2037, discusses FERC's authority to coordinate license terms of hydropower projects within a shared river basin and provides basin-wide case studies and considerations to hydropower relicensing. In the United States there are 1,043 active FERC-licensed hydroelectric projects with a total capacity of 56,097 MW. Of those, 647 with a total capacity of 21,870 MW have license expiration dates from 2018-2037. The expected workload in conjunction with the time, cost, and complexity associated with issuing a single new license has led to initiatives that aim to increase the efficiency of the relicensing process. Federal and state regulators and licensees in California, Maine, New York, and Wisconsin have begun to develop approaches to look at hydropower relicensing as part of a larger system - a river basin. These basin-wide approaches seek to coordinate different stages of the relicensing process for multiple projects at the same time. The goal of these basin-wide approaches is to increase the efficiency of the relicensing process and allow for a more comprehensive analysis of the cumulative impacts of the projects within the basin. These innovative approaches to relicensing could also reduce the workload for regulators and costs associated with relicensing. This presentation discusses some of the findings from the Basin-wide Approaches to Hydropower Relicensing: Case Studies and Considerations including a summary of the key considerations for the FERC relicensing process for non-federal hydropower projects, key findings from the analysis of FERC-licensed projects with license expiration dates from 2018-2037, discusses FERC's authority to coordinate license terms of hydropower projects within a shared river basin and provides basin-wide case studies and considerations to hydropower relicensing.

Statistical analysis of the performance and long-term stability of unquenched LSC standards ( 3 H, 14 C) used for radiochemical measurements

Multiple sets of toluene-based unquenched standards, procured over the last 3 decades, were analyzed to determine their stability over time. A statistical analysis was performed to provide insight into the variability for tSIE and counting efficiency measurements of 3 H and 14 C isotopes. Our data suggests that standards remain viable even 30 years after the manufacturer’s recommended expiration date. Overall, these standards have shown no statistically significant sign of performance degradation. We have proven through statistical analysis that these standards can provide comparable performance over time well past their manufactured expiration date.

Methods for the Automated Determination of Sustained Maximum Amplitudes in Oscillating Signals

Machine condition monitoring has been proven to reduce machine downtime and increase productivity. The state-of-the-art research uses vibration monitoring for tasks such as maintenance and tool wear prediction. A less explored aspect is how vibration monitoring might be used to monitor equipment sensitive to vibration. In a manufacturing environment, one example of where this might be needed is in monitoring the vibration of optical linear encoders used in high-precision machine tools and coordinate measuring machines. Monitoring the vibration of sensitive equipment presents a unique case for vibration monitoring because an accurate calculation of the maximum sustained vibration is needed, as opposed to extracting trends from the data. To do this, techniques for determining sustained peaks in vibration signals are needed. Here, this work fills this gap by formalizing and testing methods for determining sustained vibration amplitudes. The methods are tested on simulated signals based on experimental data. Results show that processing the signal directly with the novel Expire Timer method produces the smallest amounts of error on average under various test conditions. Additionally, this method can operate in real-time on streaming vibration data.

In-silico quantification of intersubject variability on aerosol deposition in the oral airway

The extrathoracic oral airway I s not only a major mechanical barrier for pharmaceutical aerosols to reach the lung but also a major source of variability in lung deposition. Using computational fluid dynamics, deposition of 1-30 µm particles was predicted in 11 CT-based models of the oral airways of adults (7 healthy subjects and 4 patients with chronic obstructive pulmonary disease). Simulations were performed for mouth breathing during both inspiration and expiration at two steady-state flow rates representative of resting/light activity (18 L/min) and of that typically used during aerosol inhalation from a dry powder inhaler (45 L/min). For each subject, oral deposition increased with increasing particle size and flow rate. When averaged over all subjects, there was no significant difference in oral deposition efficiency between inspiration and expiration, unlike subregional deposition that show significantly different patterns between the two breathing phases. Consistent with previous in-vitro inspiratory studies, our data showed a large intersubject variability in oral deposition. These results highlight the need for incorporating morphological variation of the upper airway in predictive models of aerosol deposition for accurate predictions of particle dosimetry in the intrathoracic region of the lung.

Inventory Management Improvement Project

The Lawrence Livermore National Laboratory (LLNL) conducts research and development for the National Nuclear Security Agency (NNSA) and its affiliates. The Polymer AM team at LLNL conducts research and development in 3D printing, specifically direct-ink-write, in accordance with LLNL and NNSA missions. The team is composed of machine operators, chemists, testing engineers, and project engineers across multiple lab spaces with one shared storage area at LLNL. The operations use a variety of different consumables and hardware to conduct research applications for LLNL and NNSA. These items are critical to performing operation tasks; if an item is out of stock, operations associated with the respective item could be suspended for weeks. As such, the Polymer AM team manages an inventory of spare consumables and hardware to ensure these items are always available. However, the current management system, an excel sheet managed by the project engineer group, is not intuitive in providing inventory information despite the high labor utilization needed to maintain the system. As such, the team is looking to improve their inventory management that can send notifications regarding inventory needs, accessible to other team members, provide all relevant information to a specific item, and store historical information for budget and operation planning. The proposed solution is a Computer Maintenance Management System (CMMS): a web-based management software that stores inventory information and provides automated notifications. The system will store information on each item including quantity in stock, technical information, supplier information, costs, lead times, and expiration date. The system has a notifications function that can send notifications to the respective team member’s when an item needs to be counted, item inventory is low, or an item is approaching their expiration date. Information for each item can be tracked over time creating data to be used for budget planning and process improvement purposes. Due to its web-based source, the system can be accessed by the respective team member viewing technical information, quantity, and purchasing information.

Quasi‐dynamic breathing model of the lung incorporating viscoelasticity of the lung tissue

Abstract We advanced a novel model to calculate viscoelastic lung compliance and airflow resistance in presence of mucus, accounting for the quasi‐linear viscoelastic stress–strain response of the parenchyma (alveoli) tissue. We adapted a continuum‐based numerical modeling approach for the lung, integrating the fluid mechanics of the airflow within individual generations of the bronchi and alveoli. The model accounts for elasticity of the deformable bronchioles, resistance to airflow due to the presence of mucus within the bronchioles, and subsequent mucus flow. Simulated quasi‐dynamic inhalation and expiration cycles were used to characterize the net compliance and resistance of the lung, considering the rheology of the mucus and viscoelastic properties of the parenchyma tissue. The structure and material properties of the lung were identified to have an important contribution to the lung compliance and airflow resistance. The secondary objective of this work was to assess whether a higher frequency and smaller volume of harmonic air flow rate compared to a normal ventilator breathing cycle enhanced mucus outflow. Results predict, lower mucus viscosity and higher excitation frequency of breathing are favorable for the flow of mucus up the bronchi tree, towards the trachea.

Quantitative methods and modeling to assess COVID–19–interrupted in vivo pharmacokinetic bioequivalence studies with two reference batches

The coronavirus disease 2019 (COVID-19) has presented unprecedented challenges to the generic drug development, including interruptions in bioequivalence (BE) studies. Per guidance published by the US Food and Drug Administration (FDA) during the COVID-19 public health emergency, any protocol changes or alternative statistical analysis plan for COVID-19-interrupted BE study should be accompanied with adequate justifications and not lead to biased equivalence determination. In this study, we used a modeling and simulation approach to assess the potential impact of study outcomes when two different batches of a Reference Standard (RS) were to be used in an in vivo pharmacokinetic BE study due to the RS expiration during the COVID-19 pandemic. Simulations were performed with hypothetical drugs under two scenarios: (1) uninterrupted study using a single batch of an RS, and (2) interrupted study using two batches of an RS. The acceptability of BE outcomes was evaluated by comparing the results obtained from interrupted studies with those from uninterrupted studies. The simulation results demonstrated that using a conventional statistical approach to evaluate BE for COVID-19-interrupted studies may be acceptable based on the pooled data from two batches. An alternative statistical method which includes a “batch” effect to the mixed effects model may be used when a significant “batch” effect was found in interrupted four-way crossover studies. However, such alternative method is not applicable for interrupted two-way crossover studies. Overall, the simulated scenarios are only for demonstration purpose, the acceptability of BE outcomes for the COVID19-interrupted studies could be case-specific.

Genomic variation within the maize stiff-stalk heterotic germplasm pool

The stiff-stalk heterotic group in Maize (Zea mays L.) is an important source of inbreds used in U.S. commercial hybrid production. Founder inbreds B14, B37, B73, and, to a lesser extent, B84, are found in the pedigrees of a majority of commercial seed parent inbred lines. We created high-quality genome assemblies of B84 and four expired Plant Variety Protection (ex-PVP) lines LH145 representing B14, NKH8431 of mixed descent, PHB47 representing B37, and PHJ40, which is a Pioneer Hi-Bred International (PHI) early stiff-stalk type. Sequence was generated using long-read sequencing achieving highly contiguous assemblies of 2.13-2.18 Gbp with N50 scaffold lengths >200 Mbp. Inbred-specific gene annotations were generated using a core five-tissue gene expression atlas, whereas transposable element (TE) annotation was conducted using de novo and homology-directed methodologies. Compared with the reference inbred B73, synteny analyses revealed extensive collinearity across the five stiff-stalk genomes, although unique components of the maize pangenome were detected. Comparison of this set of stiff-stalk inbreds with the original Iowa Stiff Stalk Synthetic breeding population revealed that these inbreds represent only a proportion of variation in the original stiff-stalk pool and there are highly conserved haplotypes in released public and ex-Plant Variety Protection inbreds. Despite the reduction in variation from the original stiff-stalk population, substantial genetic and genomic variation was identified supporting the potential for continued breeding success in this pool. The assemblies described here represent stiff-stalk inbreds that have historical and commercial relevance and provide further insight into the emerging maize pangenome.

A Prospective Trial Demonstrating the Benefit of Personalized Selection Of Breath-Hold Technique for Upper-Abdominal Radiation Therapy Using the Active Breathing Coordinator

For upper abdominal tumors, our institutional-standard motion reduction method is the expiration breath-hold (EBH) technique, using Active Breathing Coordinator (ABC). However, an individual patient's breath-hold (BH) reproducibility (R{sub BH}) may be improved in deep inspiration or inspiration breath-hold (DIBH or IBH). This trial compared the tumor position R{sub BH}, stability (S{sub BH}), and breath-hold time (T{sub BH}) of 3 BH methods, using ABC, to personalize the selection of technique, by using a preplanning screening assessment.

A framework and calculator for evaluating the impacts of shelf life extension and other food loss and waste reduction technologies

Optimization of the food supply chain (FSC) depends on reducing food waste, especially at the consumer stage, where a substantial portion of food is not eaten, but instead disposed of via landfill, incineration, or in-sink disposals. One key strategy is to increase the time that consumers have before food goes bad or expires. This study developed a framework to assess the efficacy of shelf-life extension (SLE) technologies for mitigating food loss and waste (FLW), such as packaging improvements. The impact flows through the entire FSC, reducing FLW, energy use, and other inputs at each stage. The framework and resulting calculator can be used to evaluate the impact of FLW reduction at any stage for any food commodity. As shown by two SLE cases, the calculator is valuable for policy-makers, government entities, and professionals, specifically those in marketing, business development, and capital projects teams, to comprehensively evaluate the impacts of FLW reduction technologies and practices. The framework and calculator are sensitive to the shape of the consumption curve, the fraction of inedible waste, and the current shelf life. The calculator was used to assess the impacts of the United States goal of reducing food waste by consumers through various SLE lengths. It was found that uptake of several near-ready-to-deploy SLE technologies would reduce annual food production demand by about 10–19 MMT and supply chain energy consumption by 240–410 PJ in the United States.

Quantification of SARS-CoV-2 spike protein expression from mRNA vaccines using isotope dilution mass spectrometry

The advent of mRNA vaccine technology has been vital in rapidly creating and manufacturing COVID-19 vaccines at an industrial scale. To continue to accelerate this leading vaccine technology, an accurate method is needed to quantify antigens produced by the transfection of cells with a mRNA vaccine product. This will allow monitoring of protein expression during mRNA vaccine development and provide information on how changes to vaccine components affects the expression of the desired antigen. Developing novel approaches that allow for high-throughput screening of vaccines to detect changes in antigen production in cell culture prior to in vivo studies could aid vaccine development. Here, we have developed and optimized an isotope dilution mass spectrometry method to detect and quantify the spike protein expressed after transfection of baby hamster kidney cells with expired COVID-19 mRNA vaccines. Five peptides of the spike protein are simultaneously quantified and provide assurance that protein digestion in the region of the target peptides is complete since results between the five peptides had a relative standard deviation of less than 15 %. In addition, two housekeeping proteins, actin and GAPDH, are quantified in the same analytical run to account for any variation in cell growth within the experiment. IDMS allows a precise and accurate means to quantify protein expression by mammalian cells transfected with an mRNA vaccine.

Disentangling climate and policy uncertainties for the Colorado River post-2026 operations

Abstract Lakes Mead and Powell in the Colorado River Basin underpin water and hydropower supply for the western United States. While the policies currently regulating the basin will expire by 2026, planning remains challenging due to intertwined climate variability and policy uncertainties. Based on streamflow projections from 10 dynamically downscaled CMIP6 global climate models and unique methods that add and remove internal variability, we evaluate future conditions at Powell and Mead under existing and alternative policies. Due to projected streamflow declines, under existing policy, both reservoirs will face substantial risks (>80% likelihood) of reaching dead pool before 2060. Adopting recently proposed alternative policies reduces but doesn’t eliminate such risks. All policies also exhibit tipping points where reservoir levels can change rapidly with a slight change in streamflow. A sustainable policy may require larger reductions to further reduce the reservoirs’ dead pool risks and provide better buffers from sudden changes.

Space Charge Forces analytical model for emittance compensation

Space charge forces represent main induced effects in an RF-injector that degrade the beam quality. In this scenario the laser distribution sent on the photocathode acquires an important role in the emittance compensation process, as the slice analysis shows. Starting from the preliminary studies performed on [1], a novel semi-analytical model of space charge forces is proposed in detail for bunch with arbitrary charge distribution to derive expressions of self-induced forces. The performance of the fields at low energy regime (as the field has not expired RF forces) is under present analysis, we can investigate use of this model in low charge regime. Further, the model has been bench-marked with the behavior of the distributions present in the literature and studied for new ones. It has also been applied for the study of the optimization of a C-band hybrid photoinjector now being commissioned, thus explaining the factor two reduction of the emittance observed at the exit of the gun by changing the initial distribution at the cathode.

Security Catch and Release Automation Manager (SCRAM) v1.0

This is a security automation tool heavily influenced by the tool written at NCSA https://github.com/ncsa/bhr-site. The tool is a web interface that aims to centralize information on blocking malicious traffic.
