Recombinant LAC polynucleotides and uses thereof to increase production of C-lignin in plants

The disclosure provides recombinant LAC polynucleotides encoding a laccase (LAC) polypeptide capable of polymerizing caffeyl alcohol, vectors and cells including the recombinant LAC polynucleotide. The disclosure also provides transgenic plants including cells having the recombinant LAC polynucleotide of the present disclosure and methods of increasing production of C-lignin in a plant.

Lac du Flambeau - Climate Change Resilience Initiative (Final Report)

The goals of this project were to create a comprehensive Environmental Resiliency Plan and to increase the capacity of all Lac du Flambeau Tribal Program Managers to incorporate environmental impact thinking into the everyday management of their departments. Environmental Resiliency Planning utilized current Tribal documents, which include the Integrated Resource Management Plan (IRMP), the Lac du Flambeau Tribal Emergency Management Plan (EMP), and the Lac du Flambeau Strategic Energy Plan, as guidance throughout the process. A vulnerability assessment initiated our technical process and allowed us to identify, quantify, and prioritize (or rank) the vulnerabilities in our system. This set the stage for a reduction and resilience plan as well as providing insight for revisions of existing management plans such as the Energy and Integrated Resources Management Plans

Genetic switching by the Lac repressor is based on two-state Monod–Wyman–Changeux allostery

High-resolution NMR spectroscopy enabled us to characterize allosteric transitions between various functional states of the dimeric Escherichia coli Lac repressor. In the absence of ligands, the dimer exists in a dynamic equilibrium between DNA-bound and inducer-bound conformations. Binding of either effector shifts this equilibrium toward either bound state. Analysis of the ternary complex between repressor, operator DNA, and inducer shows how adding the inducer results in allosteric changes that disrupt the interdomain contacts between the inducer binding and DNA binding domains and how this in turn leads to destabilization of the hinge helices and release of the Lac repressor from the operator. Based on our data, the allosteric mechanism of the induction process is in full agreement with the well-known Monod–Wyman–Changeux model.

Kink-driven magnetic reconnection in relativistic jets: consequences for X-ray polarimetry of BL Lacs

ABSTRACT We investigate with relativistic MHD (magnetohydrodynamic) simulations the dissipation physics of BL Lac jets, by studying the synchrotron polarization signatures of particles accelerated by the kink instability in a magnetically dominated plasma column. The non-linear stage of the kink instability generates current sheets, where particles can be efficiently accelerated via magnetic reconnection. We identify current sheets as regions where s = Jδ/B is above some predefined threshold (where B is the field strength, J is the current density, and δ is the grid scale), and assume that the particle injection efficiency scales as ∝J2. X-ray emitting particles have short cooling times, so they only probe the field geometry of their injection sites. In contrast, particles emitting in the optical band, which we follow self-consistently as they propagate away from their injection sites while cooling, sample a larger volume, and so they may be expected to produce different polarimetric signatures. We find that the degree of polarization is roughly the same between X-ray and optical bands, because even the optical emitting particles do not travel far from the current sheet where they were injected, due to lack of sufficient kink-generated turbulence. The polarization angle shows a different temporal evolution between the two bands, due to the different regions probed by X-ray and optical emitting particles. In view of the upcoming IXPE satellite, our results can help constrain whether kink-induced reconnection (as opposed to shocks) can be the source of multiwavelength emission from BL Lacs.

LAC Attachments Modernization

This document details the proposed plan to modernize the existing set of cutting attachments in use with the Liquid Abrasive Cutter (LAC). This effort will leverage the replacement of the LAC power generating unit (PGU), which is to be completed by the end of FY25. The PGU modernization will transition away from a diesel direct drive of a high pressure pump to the use of a variable frequency drive, which will be powered by electricity from the existing generator in use by our DOD customers. The existing PGU and a model of the planned replacement is shown below in Figure 1.

Flat-spectrum Radio Quasars and BL Lacs Dominate the Anisotropy of the Unresolved Gamma-Ray Background

We analyze the angular power spectrum (APS) of the unresolved gamma-ray background (UGRB) emission and combine it with the measured properties of the resolved gamma-ray sources of the Fermi-LAT 4FGL catalog. Our goals are to dissect the composition of the gamma-ray sky and to establish the relevance of different classes of source populations of active galactic nuclei in determining the observed size of the UGRB anisotropy, especially at low energies. We find that, under physical assumptions for the spectral energy distribution, i.e., by using the 4FGL catalog data as a prior, two populations are required to fit the APS data, namely flat-spectrum radio quasars at low energies and BL Lacs at higher energies. The inferred luminosity functions agree well with the extrapolation of the flat-spectrum radio quasar and BL Lac ones obtained from the 4FLG catalog. We use these luminosity functions to calculate the UGRB intensity from blazars, finding a contribution of 20% at 1 GeV and 30% above 10 GeV. Finally, bounds on an additional gamma-ray emission due to annihilating dark matter are also derived.

Materials Data on LaC by Materials Project

LaC is Tungsten Carbide structured and crystallizes in the hexagonal P-6m2 space group. The structure is three-dimensional. La3+ is bonded to six equivalent C3- atoms to form a mixture of distorted edge, face, and corner-sharing LaC6 pentagonal pyramids. All La–C bond lengths are 2.70 Å. C3- is bonded to six equivalent La3+ atoms to form a mixture of distorted edge, face, and corner-sharing CLa6 pentagonal pyramids.

The relation between optical and γ-ray emission in BL Lac sources

ABSTRACT The relativistic jets produced by some Active Galactic Nuclei (AGNs) are among the most efficient persistent sources of non-thermal radiation and represent an ideal laboratory for studying high-energy interactions. In particular, when the relativistic jet propagates along the observer’s line of sight, the beaming effect produces dominant signatures in the observed spectral energy distribution (SED), from the radio domain up to the highest energies, with the further possibility of resulting in radiation-particle multimessenger associations. In this work, we investigate the relationships between the emission of γ rays and the optical spectra of a sample of AGN, selected from BL Lac sources detected by the Fermi Large Area Telescope (Fermi-LAT). We find that there is a close relationship between the optical and γ-ray spectral indices. Despite all the limitations due to the non-simultaneity of the data, this observation strongly supports a substantial role of Synchrotron-Self Compton (SSC) radiation in a single zone leptonic scenario for most sources. This result simplifies the application of theoretical models to explore the physical parameters of the jets in this type of sources.

Emerging Modification Technologies of Lignin-based Activated Carbon toward Advanced Applications

Lignin-based activated carbon (LAC) is a promising high-quality functional material due to high surface area, abundant porous structure, and various functional groups. Modification is the most important step to functionalize LAC by altering its porous and chemical properties. Here, this Review summarizes the state-of-the-art modification technologies of LAC toward advanced applications. Promising modification approaches are reviewed to display their effects on the preparation of LAC. The multiscale changes in the porosity and the surface chemistry of LAC are fully discussed. Advanced applications are then introduced to show the potential of LAC for supercapacitor electrode, catalyst support, hydrogen storage, and carbon dioxide capture. Finally, the mechanistic structure-function relationships of LAC are elaborated. These results highlight that modification technologies play a special role in altering the properties and defining the functionalities of LAC, which could be a promising porous carbon material toward industrial applications.

Compensatory reserve detects subclinical shock with more expeditious prediction for need of life‐saving interventions compared to systolic blood pressure and blood lactate

Abstract Introduction We conducted a prospective observational study on 205 trauma patients at a level I trauma facility to test the hypothesis that a compensatory reserve measurement (CRM) would identify higher risk for progression to shock and/or need a life‐saving interventions (LSIs) earlier than systolic blood pressure (SBP) and blood lactate (LAC). Methods A composite outcome metric included blood transfusion, procedural LSI, and mortality. Discrete measures assessed as abnormal (ab) were SBP <90 mmHg, CRM <60%, and LAC >2.0. A graded categorization of shock was defined as: no shock (normal [n] SBP [n‐SBP], n‐CRM, n‐LAC); sub‐clinical shock (ab‐CRM, n‐SBP, n‐LAC); occult shock (n‐SBP, ab‐CRM, ab‐LAC); or overt shock (ab‐SBP, ab‐CRM, ab‐LAC). Results Three patients displayed overt shock, 53 displayed sub‐clinical shock, and 149 displayed no shock. After incorporating lactate into the analysis, 86 patients demonstrated no shock, 25 were classified as sub‐clinical shock, 91 were classified as occult shock, and 3 were characterized as overt shock. Each shock subcategory revealed a graded increase requiring LSI and transfusion. Initial CRM was associated with progression to shock (odds ratio = 0.97; p < .001) at an earlier time than SBP or LAC. Conclusions Initial CRM uncovers a clinically relevant subset of patients who are not detected by SBP and LAC. Our results suggest CRM could be used to more expeditiously identify injured patients likely to deteriorate to shock, with requirements for blood transfusion or procedural LSI.

Drivers of Future Physical Water Scarcity and Its Economic Impacts in Latin America and the Caribbean

Future water scarcity is a global concern with significant impacts on the energy, water, and land (EWL) sectors. Countries in Latin America and the Caribbean (LAC) are significant producers of agricultural goods consumed around the world, so disruptions resulting from land and water scarcity in LAC are a source of global supply chain risk. Understanding where water scarcity in LAC could occur and what could drive that scarcity to emerge is critical for strategic resource management and planning, both regionally and globally. Assessing future water scarcity impacts in LAC is challenging given the multisector dynamic interactions among the EWL sectors and the multiple uncertainties acting across different spatial scales. We use scenario discovery to explore the drivers of future water scarcity, considering assumptions related to climate, socioeconomics, and the EWL sectors. Understanding what factors drive uncertainty across outcomes can help stakeholders in targeting long-term solutions and future data collection efforts. To illuminate these dynamics, we use a database consisting of a large ensemble of scenarios representing diverse worlds simulated using the Global Change Analysis Model. We quantify future water scarcity and its economic impacts across the scenarios using three metrics: (1) physical water scarcity, (2) water price, and (3) crop profit. We find that physical water scarcity and water price are driven primarily by reservoir storage capacity assumptions, highlighting the importance of strategic water infrastructure development in maintaining future water availability and accessibility. Crop profit is driven by a combination of water supply and demand assumptions, which emphasizes the complex nature of EWL multisector dynamics. While most of LAC is poised to have abundant land and water resources available for development, water basins in Mexico and along the Pacific coast of South America experience high levels of severity and uncertainty across the future scenarios for at least one of the metrics. We find that the drivers of extreme scarcity vary spatially and across the metrics, further highlighting the heterogeneity of the region and the importance of considering multiple metrics to assess water scarcity vulnerability.

Multiplexed CRISPR/Cas9 editing of the long-chain acyl-CoA synthetase family in the diatom Phaeodactylum tricornutum reveals that mitochondrial ptACSL3 is involved in the synthesis of storage lipids

Long-chain acyl-CoA synthetases (LACS) play diverse and fundamentally important roles in lipid metabolism. While their functions have been well established in bacteria, yeast and plants, the mechanisms by which LACS isozymes regulate lipid metabolism in unicellular oil-producing microalgae, including the diatom Phaeodactylum tricornutum , remain largely unknown. In P. tricornutum , a family of five genes ( ptACSL1 – ptACSL5 ) encodes LACS activities. For this work, we generated single lacs knockout/knockdown mutants using multiplexed CRISPR/Cas9 method, and determined their substrate specificities towards different fatty acids (FAs) and subcellular localisations. ptACSL3 is localised in the mitochondria and its disruption led to compromised growth and reduced triacylglycerol (TAG) content when cells were bubbled with air. The ptACSL3 mutants showed altered FA profiles in two galactoglycerolipids and phosphatidylcholine (PC) with significantly reduced distribution of 16:0 and 16:1. ptACSL5 is localised in the peroxisome and its knockdown resulted in reduced growth rate and altered molecular species of PC and TAG, indicating a role in controlling the composition of acyl-CoAs for lipid synthesis. Our work demonstrates the potential of generating gene knockout mutants with the mutation of large fragment deletion using multiplexed CRISPR/Cas9 and provides insight into the functions of LACS isozymes in lipid metabolism in the oleaginous microalgae.

An Engineered Laccase from Fomitiporia mediterranea Accelerates Lignocellulose Degradation

Laccases from white-rot fungi catalyze lignin depolymerization, a critical first step to upgrading lignin to valuable biodiesel fuels and chemicals. In this study, a wildtype laccase from the basidiomycete Fomitiporia mediterranea (Fom_lac) and a variant engineered to have a carbohydrate-binding module (Fom_CBM) were studied for their ability to catalyze cleavage of β-O-4′ ether and C–C bonds in phenolic and non-phenolic lignin dimers using a nanostructure-initiator mass spectrometry-based assay. Fom_lac and Fom_CBM catalyze β-O-4′ ether and C–C bond breaking, with higher activity under acidic conditions (pH < 6). The potential of Fom_lac and Fom_CBM to enhance saccharification yields from untreated and ionic liquid pretreated pine was also investigated. Adding Fom_CBM to mixtures of cellulases and hemicellulases improved sugar yields by 140% on untreated pine and 32% on cholinium lysinate pretreated pine when compared to the inclusion of Fom_lac to the same mixtures. Adding either Fom_lac or Fom_CBM to mixtures of cellulases and hemicellulases effectively accelerates enzymatic hydrolysis, demonstrating its potential applications for lignocellulose valorization. We postulate that additional increases in sugar yields for the Fom_CBM enzyme mixtures were due to Fom_CBM being brought more proximal to lignin through binding to either cellulose or lignin itself.

K2 Optical Emission from OJ 287 and Other γ-Ray Blazars on Hours-to-weeks Timescales from 2014 to 2018

We present second observations by K2 of OJ 287 and seven other γ-ray active galactic nuclei (AGNs) obtained in 2017–2018, second and third observations of the lobe-dominated, steep-spectrum quasar 3C 207, and observations of nine additional blazars not previously observed with K2. The AGNs were observed simultaneously with K2 and the Fermi Large Area Telescope for 51–81 days. Our full sample, observed in 2014–2018, contained 16 BL Lacertae objects (BL Lacs), nine flat-spectrum radio quasars (FSRQs), and four other γ-ray AGNs. Twelve BL Lacs and seven FSRQs exhibited fast, jagged light curves, while four BL Lacs and two FSRQs had slow, smooth light curves. Some objects changed their red-noise character significantly between repeated K2 observations. The optical characteristics of OJ 287 derived from the short-cadence K2 light curves changed between observations made before and after the predicted passage of the suspected secondary supermassive black hole through the accretion disk of the primary supermassive black hole. The average slopes of the periodogram power spectral densities of the BL Lacs' and FSRQs' light curves differed significantly, by ≈12%, with the BL Lac slopes being steeper, and a Kolmogorov–Smirnov test with a p-value of 0.039 indicates that these samples probably come from different populations; however, this result is not as strongly supported by power spectral response (PSRESP) analyses. Differences in the origin of the jets from the ergosphere or accretion disk in these two classes could produce such a disparity, as could different sizes or locations of emission regions within the jets.

SNICAR-ADv4: a physically based radiative transfer model to represent the spectral albedo of glacier ice

Abstract. Accurate modeling of cryospheric surface albedo is essential for our understanding of climate change as snow and ice surfaces regulate the global radiative budget and sea-level through their albedo and mass balance. Although significant progress has been made using physical principles to represent the dynamic albedo of snow, models of glacier ice albedo tend to be heavily parameterized and not explicitly connected with physical properties that govern albedo, such as the number and size of air bubbles, specific surface area (SSA), presence of abiotic and biotic light absorbing constituents (LACs), and characteristics of any overlying snow. Here, we introduce SNICAR-ADv4, an extension of the multi-layer two-stream delta-Eddington radiative transfer model with the adding–doubling solver that has been previously applied to represent snow and sea-ice spectral albedo. SNICAR-ADv4 treats spectrally resolved Fresnel reflectance and transmittance between overlying snow and higher-density glacier ice, scattering by air bubbles of varying sizes, and numerous types of LACs. SNICAR-ADv4 simulates a wide range of clean snow and ice broadband albedo (BBA), ranging from 0.88 for (30 µm) fine-grain snow to 0.03 for bare and bubble-free ice under direct light. Our results indicate that representing ice with a density of 650 kg m−3 as snow with no refractive Fresnel layer, as done previously, generally overestimates the BBA by an average of 0.058. However, because most naturally occurring ice surfaces are roughened “white ice”, we recommend modeling a thin snow layer over bare ice simulations. We find optimal agreement with measurements by representing cryospheric media with densities less than 650 kg m−3 as snow and larger-density media as bubbly ice with a Fresnel layer. SNICAR-ADv4 also simulates the non-linear albedo impacts from LACs with changing ice SSA, with peak impact per unit mass of LACs near SSAs of 0.1–0.01 m2 kg−1. For bare, bubble-free ice, LACs actually increase the albedo. SNICAR-ADv4 represents smooth transitions between snow, firn, and ice surfaces and accurately reproduces measured spectral albedos of a variety of glacier surfaces. This work paves the way for adapting SNICAR-ADv4 to be used in land ice model components of Earth system models.

A lipase from Lacticaseibacillus rhamnosus IDCC 3201 with thermostability and pH resistance for use as a detergent additive

Lipases are important biocatalysts and ubiquitous in plants, animals, and microorganisms. The high growth rates of microorganisms with low production costs have enabled the wide application of microbial lipases in detergent, food, and cosmetic industries. Herein, a novel lipase from Lacticaseibacillus rhamnosus IDCC 3201 (Lac-Rh) was isolated and its activity analyzed under a range of reaction conditions to evaluate its potential industrial application. The isolated Lac-Rh showed a molecular weight of 24 kDa and a maximum activity of 3438.5 ± 1.8 U/mg protein at 60 °C and pH 8. Additionally, Lac-Rh retained activity in alkaline conditions and in 10% v/v concentrations of organic solvents, including glycerol and acetone. Interestingly, after pre-incubation in the presence of multiple commercial detergents, Lac-Rh maintained over 80% of its activity and the stains from cotton were successfully removed under a simulated laundry setting. Overall, the purified lipase from L. rhamnosus IDCC 3201 has potential for use as a detergent in industrial applications.
