A nut-and-bolt assembly of the bimodular large progenitor botulinum neurotoxin complex

Botulinum neurotoxin serotype A (BoNT/A) is naturally produced by bacteria along with four nontoxic neurotoxin-associated proteins (NTNH, HA70, HA33, and HA17), forming a bimodular large progenitor toxin complex (L-PTC). The BoNT/A–NTNH complex protects the toxin from adverse environment, while the complex consisting of HA proteins facilitates toxin absorption during oral intoxication. How these two independent modules assemble into the L-PTC remains unclear. Here, we report the crystal structure of the BoNT/A–NTNH–HA70 complex at ~2.9-Å resolution. The structure reveals that the BoNT/A–NTNH complex is anchored into a concentric double β-barrel channel of trimeric HA70 through a short β-hairpin of NTNH (termed nLoop), resembling a nut-and-bolt attachment. We find that the nLoop of NTNH is strictly conserved across HA-containing BoNT complexes and that NTNH–HA70 binding is interchangeable among them. Furthermore, we demonstrate that the nLoop functions as a minimal motif enabling attachment of a protein-of-interest to the HA complex, with potential applications in oral biologics delivery.

MPI nuts and bolts and more [Slides]

MPI (Message-Passing Interface) is a message-passing library interface specification. All parts of this definition are significant. MPI addresses primarily the message-passing parallel programming model, in which data is moved from the address space of one process to that of another process through cooperative operations on each process. . . MPI is a specification, not an implementation; there are multiple implementations of MPI. This specification is for a library interface; MPI is not a language, and all MPI operations are expressed as functions, subroutines, or methods, according to the appropriate language bindings that, for C and Fortran, are part of the MPI standard. MPI Forum is the organization which is responsible for the MPI Specification.

Resilient Solar PV Structures a.k.a. the "Nuts and Bolts" of PV Resilience

Solar photovoltaic (PV) systems can and has survived many extreme weather events. On occasion, however, solar PV systems have been damaged. This damage is related more to the features of individual PV systems rather than the magnitude of the weather events. Much of this damage can be avoided with relatively simple, cost-effective measures that include design, installation, inspection, and O&M best practices.

Maturing Rational Design Methodologies and Industry Consensus Engineering Standards: Critical Fastened Joints - Solar PV Industry

Critical structural joints can be seen throughout a solar array and are called upon to secure modules and keep racking assembled and able to resist large demands from winds and snow loads. In the relatively new and fast-growing solar PV industry, the important role these hardware assemblies (e.g. clips, clamps, bolts, nuts, washers) play is not well understood by product designers. Failures with critical structural joints are surprisingly common and point to the need for maturing the engineering and assembly of these joints. The wide variety of design concepts (Figure 2&2) demonstrate interesting and innovative ideas but are lacking the basics of fastener engineering seen in matured industries (e.g. transportation, buildings). Complicating the maturing process for critical structural joints is that they are one component in rack supporting structures that exhibits a systems behavior; each component will affect the other and play a key role in maintaining structural integrity. When wind loads the surface of a module, the underlying racking members deflect and twist which in turn imparts forces back into the joints and into the mounted modules. Often, these supporting rack structures exhibit high deflections and low natural frequencies which amplify the demands placed into the joints even in moderate winds. Current engineering practices and associated structural conventions view solar racking support structures as they would a high mass building that exhibit more static behaviors in wind events. Solar structures are unique from high mass buildings and require the development of solar specific industry engineering consensus standards.

Idealized Simulations of the Tropical Climate and Variability in the Single Column Atmosphere Model (SCAM): Radiative-Convective Equilibrium

To explore the interactions among column processes in the Community Atmosphere Model (CAM), the single-column version of CAM (SCAM) is integrated for 1000 days in radiative-convective equilibrium (RCE) with tropical values of boundary conditions, spanning a parameter or configuration space of model physics versions (v5 vs. v6), vertical resolution (standard and 60 levels), sea surface temperature (SST), and some interpretation-driven experiments. The simulated time-mean climate is reasonable, near observations and RCE of a cyclic cloud-resolving model. Updraft detrainment in the deep convection scheme produces distinctive grid-scale structures in humidity and cloud, which also interact with radiative transfer processes. These grid artifacts average out in multi-column RCE results reported elsewhere, illustrating the nuts-and-bolts interpretability that SCAM adds to the hierarchy of model configurations. Multi-day oscillations of precipitation arise from descent of warm convection-capping layers starting near the tropopause, eventually reset by a burst of convective deepening. Experiments reveal how these oscillations depend critically on an internal parameter that controls the number of neutral buoyancy levels allowed for determining cloud top and computing dilute convective available potential energy in the deep convection scheme, and merely modified a little by disabling cloud-base radiation (heating of cloud base). This strong dependence of transient behavior in 1D on this parameter will be tested in the second part of this work, in which SCAM is coupled to a parameterized dynamics of two-dimensional, linearized gravity wave, and in the 3D simulations in future study.

A framework for materials informatics education through workshops

The burgeoning field of materials informatics necessitates a focus on educating the next generation of materials scientists in the concepts of data science, artificial intelligence (AI), and machine learning (ML). In addition to incorporating these topics in undergraduate and graduate curricula, regular hands-on workshops present the most effective medium to initiate researchers to informatics and have them start applying the best AI/ML tools to their own research. With the help of the Materials Research Society (MRS), members of the MRS AI Staging Committee, and a dedicated team of instructors, we successfully conducted workshops covering the essential concepts of AI/ML as applied to materials data, at both the Spring and Fall Meetings in 2022, with plans to make this a regular feature in future meetings. Here, in this article, we discuss the importance of materials informatics education via the lens of these workshops, including details such as learning and implementing specific algorithms, the crucial nuts and bolts of ML, and using competitions to increase interest and participation.

Complement to thermodynamics of dyonic Taub-NUT-AdS spacetime

We examine the thermodynamics of Euclidean dyonic Taub-Nut/Bolt-AdS 4 black holes for a variety of horizon geometries to understand how gauge field regularity conditions influence the thermodynamic relations. We find several distinct features that distinguish the NUT-charged case from its dyonic Reissner-Nordstrom counterpart. For the Nut solution, the gauge field vanishes at the horizon and so regularity is ensured. For the Bolt solution we find that the norm of the gauge field is required to vanish at the horizon in order to satisfy both regularity and the first law of thermodynamics. This regularity condition yields a constraint on the electric and magnetic charges and so reduces cohomogeneity of the system; for spherical horizons, the regularity condition removing the Misner string singularity further reduces cohomogeneity. We observe that bolt solutions with increasing electric charge have positive heat capacity, but upon turning on the magnetic charge to make the solution dyonic, we find that the properties of the uncharged one are retained, having both positive and negative heat capacity. We also study the extremal Bolt solution, finding that Misner string disappears at the horizon in the zero temperature limit. We find that the extremal solution has finite-temperature-like behaviour, with the electric potential playing a role similar to temperature.
