Modified biosensors and biocatalysts and methods of use

Biosensors including a nucleic acid encoding a PcaU protein, a PobR protein, a CatM protein, a PcaR protein, or a TphR protein are provided. In some examples, the biosensors include a promoter regulated by the sensed molecule operably linked to a reporter gene. The biosensors may be included in a vector or in cells including one or more of the biosensors or vectors. Modified chorismate pyruvate lyase (UbiC) and modified paraoxonase (PON1) proteins including one or more amino acid substitutions are provided. Finally, methods of selecting biocatalysts with increased activity including transforming a library of cells expressing a biosensor with one or more nucleic acids encoding one or more mutations in a gene involved in a biosynthesis pathway, determining activity of the reporter protein; and selecting a cell with increased reporter protein activity as expressing a biocatalyst with increased activity are provided.

ECAR-6330 Rev 0 Computational Fluid Dynamics Evaluation of Core Pressure Drop for the Primary Coolant Apparatus Test

PCAT is a thermal-hydraulic test loop for MARVEL reactor. The design gap thickness between the heater rods of PCAT core is 1.0 mm. The heater elements were manufactured within a straightness tolerance for the designed gap thickness, but several elements have been distorted out of tolerance during the welding process. The distortion of heater rods could lead to flow maldistribution and excessive hotspot temperature when the gap between two heater rods is too narrow. Wire-wraps are the spacer devices widely used in the sodium cooled fast reactor system to maintain the design fuel pitch. Generally, a wire spacer can enhance the turbulent heat transfer, but it increases the pressure drop, which is not desirable for a natural-circulation system like PCAT. Therefore, three short wire-wrap spacers per heater have been proposed to contain the core pressure drops and keep the heater-to-heater gap in an acceptable range. The objectives of this ECAR is to evaluate the PCAR core pressure drop with short wire spacers to confirm that the additional pressure drop due to the wire spacer will not disturb the natural circulation of the PCAT.

Noncontact Andreev Reflection as a Direct Probe of Superconductivity on the Atomic Scale

Direct detection of superconductivity has long been a key strength of point-contact Andreev reflection. However, its applicability to atomic-scale imaging is limited by the mechanical contact of the Andreev probe. To this end, we present a new method to probe Andreev reflection in a tunnel junction, leveraging tunneling spectroscopy and junction tunability to achieve quantitative detection of Andreev scattering. This method enables unambiguous assignment of superconducting origins of current-carrying excitations, as well as detection of higher order Andreev processes in atomic-scale junctions. Herein, we furthermore revealed distinct sensitivity of Andreev reflection to natural defects, such as step edges, even in classical superconductors. The methodology opens a new path to nano- and atomic-scale imaging of superconducting properties, including disordered superconductors and proximity to phase transitions.
