CD8 Lymphocyte Depletion Enhances the Latency Reversal Activity of the SMAC Mimetic AZD5582 in ART-Suppressed Simian Immunodeficiency Virus-Infected Rhesus Macaques

ABSTRACT Inducing latency reversal to reveal infected cells to the host immune system represents a potential strategy to cure human immunodeficiency virus (HIV) infection. In separate studies, we have shown previously that CD8 + T cells may contribute to the maintenance of viral latency and have identified a novel SMAC (second mitochondrial activator of caspases) mimetic/IAP (inhibitor of apoptosis protein) inhibitor (AZD5582) capable of reversing HIV/simian immunodeficiency virus (SIV) latency in vivo by activating the noncanonical (nc) NF-κB pathway. Here, we use AZD5582 in combination with antibody (Ab)-mediated depletion of CD8α + cells to further evaluate the role of CD8 + T cells in the maintenance of viral latency. Six rhesus macaques (RMs) were infected with SIV mac239 and treated with antiretroviral therapy (ART) starting at week 8 postinfection. After 84 to 85 weeks of ART, all animals received a single dose of the anti-CD8α depleting Ab MT807R1 (50 mg/kg of body weight, administered subcutaneously [s.c.]), followed by five weekly doses of AZD5582 (0.1 mg/kg, administered intravenously [i.v.]). Following combined treatment with CD8α depletion and AZD5582, 100% of RMs experienced on-ART viremia levels above 60 copies per ml of plasma. In comparator groups of SIV-infected, ART-suppressed RMs treated with AZD5582 only or CD8α depletion only, on-ART viremia was experienced by 56% and 57% of the animals, respectively. Furthermore, the increased frequency of viremic episodes during the treatment period was greater in the group treated with CD8α depletion plus AZD5582 than in the other groups. Mathematical modeling of virus reactivation suggested that in addition to viral dynamics during acute infection, CD8α depletion influenced the response to AZD5582. This work suggests that the latency reversal induced by activation of the ncNF-κB signaling pathway with AZD5582 can be enhanced by CD8α + cell depletion. IMPORTANCE A favored approach to curing HIV infection aims at inducing viral expression using latency-reversing agents (LRAs) to allow the elimination of infected cells. Here, we tested a combination of two recently identified LRAs, the SMAC mimetic/IAP inhibitor AZD5582, which activates the noncanonical NF-κB pathway, and the antibody (Ab) MT807R1, which depletes CD8α + cells, in SIV-infected rhesus macaques (RMs) on ART. Latency reversal, as defined by increased on-ART viremia, was observed in all six SIV-infected, ART-treated RMs that received this combined treatment. Furthermore, comparison of viral reactivation between these animals and groups of SIV-infected, ART-treated RMs treated with AZD5582 only or CD8α + cell depletion only showed more frequent increases in viremic episodes when the two treatments were combined. This study provides additional evidence that CD8 + T cells may contribute to the maintenance of HIV/SIV latency on ART and potentially inhibit latency reversal during HIV cure approaches.

High-z galaxies with JWST and local analogues – it is not only star formation

ABSTRACT I present an analysis of the JWST NIRSpec data of SMACS 0723 released as Early Release Observations. As part of this three new redshifts are provided, bringing the total of reliable redshifts to 14. I propose a modification to the direct abundance determination method that reduces sensitivity to flux calibration uncertainties by a factor of ∼3 and show that the resulting abundances are in good agreement with Bayesian photoionization models of the rest-frame optical spectrum. I also show that 6355 is most likely a narrow-line active galactic nucleus (AGN) with $M_*\lt 10^9\, \mathrm{M}_\odot$ at z = 7.66 and argue that 10 612 might also have an AGN contribution to its flux through comparison to photoionization models and low-redshift analogues. Under the assumption that the lines come from star formation, I find that the galaxies have gas depletion times of ∼107 yr, comparable to similar galaxies locally. I also identify a population of possibly shock-dominated galaxies at z < 3, whose near-IR emission lines plausibly come nearly all from shocks and discuss their implications. I close with a discussion of the potential for biases in the determination of the mass–metallicity relation using samples defined by detected [O iii]4363 and show using low-z galaxies that this can lead to biases of up to 0.5 dex with a systematic trend with mass.

Detecting low-mass perturbers in cluster lenses using curved arc bases

ABSTRACT Strong gravitationally lensed arcs produced by galaxy clusters have been observationally detected for several decades now. These strong lensing constraints provided high-fidelity mass models for cluster lenses that include substructure down to $10^{9{-}10}\, \mathrm{M}_\odot$. Optimizing lens models, where the cluster mass distribution is modelled by a smooth component and subhaloes associated with the locations of individual cluster galaxies, has enabled deriving the subhalo mass function, providing important constraints on the nature and granularity of dark matter. In this work, we explore and present a novel method to detect and measure individual perturbers (subhaloes, line-of-sight haloes, and wandering supermassive black holes) by exploiting their proximity to highly distorted lensed arcs in galaxy clusters, and by modelling the local lensing distortions with curved arc bases. This method offers the possibility of detecting individual low-mass perturber subhaloes in clusters and haloes along the line of sight down to a mass resolution of $10^8\, \mathrm{M}_\odot$. We quantify our sensitivity to low-mass perturbers ($M\sim 10^{7{-}9}\, \mathrm{M}_\odot$) in clusters (M ∼ 1014–15 M⊙), by creating realistic mock data. Using three lensed images of a background galaxy in the cluster SMACS J0723, taken by JWST, we study the retrieval of the properties of potential perturbers with masses $M=10^{7{-}9}\, \mathrm{M}_\odot$. From the derived posterior probability distributions for the perturber, we constrain its concentration, redshift, and ellipticity. By allowing us to probe lower mass substructures, the use of curved arc bases can lead to powerful constraints on the nature of dark matter as discrimination between dark matter models appears on smaller scales.

Image Deconvolution and Point-spread Function Reconstruction with STARRED: A Wavelet-based Two-channel Method Optimized for Light-curve Extraction

We present starred, a point-spread function (PSF) reconstruction, two-channel deconvolution, and light-curve extraction method designed for high-precision photometric measurements in imaging time series. An improved resolution of the data is targeted rather than an infinite one, thereby minimizing deconvolution artifacts. In addition, starred performs a joint deconvolution of all available data, accounting for epoch-to-epoch variations of the PSF and decomposing the resulting deconvolved image into a point source and an extended source channel. The output is a high-signal-to-noise-ratio, high-resolution frame combining all data and the photometry of all point sources in the field of view as a function of time. Of note, starred also provides exquisite PSF models for each data frame. We showcase three applications of starred in the context of the imminent LSST survey and of JWST imaging: (i) the extraction of supernovae light curves and the scene representation of their host galaxy; (ii) the extraction of lensed quasar light curves for time-delay cosmography; and (iii) the measurement of the spectral energy distribution of globular clusters in the "Sparkler," a galaxy at redshift z = 1.378 strongly lensed by the galaxy cluster SMACS J0723.3-7327. starred is implemented in jax, leveraging automatic differentiation and graphics processing unit acceleration. This enables the rapid processing of large time-domain data sets, positioning the method as a powerful tool for extracting light curves from the multitude of lensed or unlensed variable and transient objects in the Rubin-LSST data, even when blended with intervening objects.
