Anomalous temperature dependence of the experimental x-ray structure factor of supercooled water

The structural changes of water upon deep supercooling were studied through wide-angle x-ray scattering at SwissFEL. The experimental setup had a momentum transfer range of 4.5 Å-1, which covered the principal doublet of the x-ray structure factor of water. The oxygen–oxygen structure factor was obtained for temperatures down to 228.5 ± 0.6 K. Similar to previous studies, the second diffraction peak increased strongly in amplitude as the structural change accelerated toward a local tetrahedral structure upon deep supercooling. We also observed an anomalous trend for the second peak position of the oxygen–oxygen structure factor (q 2 ). We found that q 2 exhibits an unprecedented positive partial derivative with respect to temperature for temperatures below 236 K. Based on Fourier inversion of our experimental data combined with reference data, we propose that the anomalous q 2 shift originates from that a repeat spacing in the tetrahedral network, associated with all peaks in the oxygen–oxygen pair-correlation function, gives rise to a less dense local ordering that resembles that of low-density amorphous ice. The findings are consistent with that liquid water consists of a pentamer-based hydrogen-bonded network with low density upon deep supercooling.

Two-color x-ray free-electron laser by photocathode laser emittance spoiler

Multispectral x-ray pump-probe experiments call for synchronized two-color free-electron lasers (FEL). This mode often implies a laborious setup or an inefficient use of the undulator. We report on a simple and noninvasive approach tested at SwissFEL for a two-color x-ray delivering almost 60% of the pulse energy compared with a single color. In this new method, a ps UV pulse is overlapped to the photocathode drive laser increasing the beam emittance, which locally inhibits the FEL process. This scheme permits high-stability in energy and spectrum and the control of the two-color duration and intensity ratio. It enables shot-to-shot switching between one and two-color FEL and, since not associate to beam losses, it is compatible with high repetition-rate and high average-power FELs.

Integrated structural dynamics uncover new modes of B12 photoreceptor activation

**SACLA** A crystallographic pump-power titration was first carried-out with pump laser fluences of 12, 30, 60 and 120 uJ.cm-2 and a time delay of 3 us. Then, a time-series was performed with a pump-laser fluence of 30 uJ.cm-2 (~2.4 absorbed photon per chromophore) and time-delays of 10 ns, 300 ns, 3 µs, 100 us and 3 ms. Finally, two time-delays (10 ns and 3 us) were collected with a pump laser fluence of 12 uJ.cm-2. Raw images, crystFEL streams and merged mtzs are available for all collected dat **SwissFEL** A time series with a pump laser fluence of 30 mJ.cm-2 was performed at time delys of 3 µs and 10 ms. X-ray Absorption Spectroscopy of Dilute Metalloenzymes at X-ray Free-Electron Lasers in a Shot-by-Shot Mode

X-ray absorption spectroscopy (XAS) of 3d transition metals provides important electronic structure information for many fields. However, X-ray-induced radiation damage under physiological temperature has prevented using this method to study dilute aqueous systems, such as metalloenzymes, as the catalytic reaction proceeds. Here we present a new approach to enable operando XAS of dilute biological samples and demonstrate its feasibility with K-edge XAS spectra from the Mn cluster in photosystem II and the Fe–S centers in photosystem I. This approach combines highly efficient sample delivery strategies and a robust signal normalization method with high-transmission Bragg diffraction-based spectrometers at X-ray free-electron lasers (XFELs) in a damage-free, shot-by-shot mode. These photon-out spectrometers have been optimized for discriminating the metal Mn/Fe Kα fluorescence signals from the overwhelming scattering background present on currently available detectors for XFELs that lack suitable energy discrimination. We quantify the enhanced performance metrics of the spectrometer and discuss its potential applications for acquiring time-resolved XAS spectra of biological samples during their reactions at XFELs.
