Pr(H 3 BNMe 2 BH 3 ) 3 and Pr(thd) 3 as Volatile Carriers for Actinium-225. Deposition of Actinium-Doped Praseodymium Boride Thin Films for Potential Use in Brachytherapy

Here we show that the praseodymium N,N-dimethylaminodiboranate complex Pr(H 3 BNMe 2 BH 3 ) 3 and the 2,2,6,6-tetramethylheptane-3,5-dionate complex Pr(thd) 3 can serve as volatile carriers for 225 Ac. The actinium coordination complexes Ac(H 3 BNMe 2 BH 3 ) 3 and Ac(thd) 3 are the likely species subliming with the carrier material. A sample of 225 Ac-doped Pr(H 3 BNMe 2 BH 3 ) 3 was used to deposit amorphous 225 Ac-doped praseodymium boride films on glass and Si(100) at 300 °C. Furthermore, the α emission spectra of the refractory films are well-resolved, suggesting that they could be used as radioactive implants for brachytherapy and related treatments.

Actinium purification from elemental components of stainless steel and aluminum

Radioisotopes of actinium are valuable because of their potential use in the medical industry. Actinium-225 shows promise for treating disease—like cancer—via targeted alpha therapy (TAT), and the longer-lived actinium-227 is the parent of two radionuclides with potential application in TAT radiopharmaceuticals, thorium-227 and radium-223. Continuing progress in the development of these medical applications requires robust and diverse methods for extracting and purifying actinium from a wide range of matrices. Further, to define the strengths and limitations of separation methods commonly employed in actinium processing, we characterized the performance of ion exchange and extraction chromatographic steps for removing contaminants found in stainless steel (chromium, iron, nickel, and silicon) and aluminum from actinium-227. While AG-MP1 anion exchange resin with HCl (aq) successfully removed iron, cation exchanger AG 50W-X8 removed most other contaminants. The most persistent contaminant was aluminum, which was removed using a DGA Normal extraction resin. These results are presented within the context of applying that methodology toward actinium purification strategies.

Accelerating actinium-225 purification by high-pressure ion chromatography

Actinium-225 (t1/2 = 9.92 days) is an important radioisotope for targeted alpha therapy applications. The limited supply obtained through the decay of thorium-229 has motivated accelerator-based production routes, including irradiation of thorium targets. Irradiated targets can produce useful quantities of actinium-225, but the product requires final purification from chemically similar lanthanide contaminants. This work describes an automated high-pressure ion chromatography method for this final polishing step. The method uses a reusable strong-acid cation-exchange column bearing sulfonic acid functional groups. α-Hydroxyisobutyric acid (α-HIBA), adjusted to pH 4.3 with lithium hydroxide, complexes and elutes lanthanides, a dilute hydrochloric acid matrix-exchange step removes residual α-HIBA, and concentrated hydrochloric acid then elutes retained actinium(III). The protocol purified actinium-225 to >99% radiopurity across tracer-level samples and samples containing >150 µCi (5.6 MBq) of activity. A 10 min, 0.1 M hydrochloric acid matrix exchange substantially reduced organic eluent carryover, and in-line sodium iodide detection enabled real-time monitoring of actinium and lanthanide elution. The developed method can be completed in <1 h and provides a basis for automated purification workflows for accelerator-produced actinium-225.

Extraction of radium and actinium with Pb resin and Rose Bengal

The extraction of radium and actinium with Eichrom Pb resin and Rose Bengal has been studied. Radium and actinium both show a strong affinity (k’ > 10,000) for the resin at intermediate pHs (~ 5–7) from solutions with Rose Bengal. The pH range and magnitude of uptake for both radium and actinium is significantly increased by the presence of Rose Bengal compared to basic solutions without this counter ion. Additionally, radium uptake is extremely fast from basic solutions with and without Rose Bengal; actinium uptake is slower, particularly in solutions without Rose Bengal. Column studies show that despite the similar behavior of actinium and radium in this system, separations of these two elements with Pb resin and Rose Bengal solutions are possible with reasonable yields due to the differences in uptake kinetics.

Extraction of radium and actinium with dibenzo-21-crown-7 resin and Rose Bengal

The extraction of 224 Ra and 228 Ac with dibenzo-21-crown-7 resin was studied in solutions with Rose Bengal and identical solutions without Rose Bengal (blanks) from pH 3 to 12. Batch studies demonstrated that actinium is extracted at high pH (> 10) from both solutions but with a significantly higher extraction from the Rose Bengal solutions (maximum extraction ~ 60,000) than the blank solutions (maximum extraction ~ 2,000). In the Rose Bengal solutions, there was a weak extraction of actinium at lower pH (4–10) as well. There was no uptake of radium from the blank solutions, and a weak extraction at all pH levels from the Rose Bengal solutions. Kinetics studies were performed for actinium only, as radium had no significant extraction. Two column studies are demonstrated for the separation of radium from actinium. These separations are moderately effective with ~ 78% of the actinium recovered with no detectable radium.

The coordination properties and ionic radius of actinium: A 120-year-old enigma

Actinium is an elusive element with untamed properties and represents a peculiar case in the periodic table, as its isotopes are all radioactive, the longest-lived one having only a 22-year half-life, and the availability of actinium isotopes remains very low (microgram level, at best), hindering research on its compounds. Despite being a natural element discovered more than 120 years ago, and despite an increasing interest in using one of its isotopes ( 225 Ac) for highly efficient cancer therapies, the chemistry of actinium is still largely unknown relative to other elements. Since Ac is the first element of the actinide series, it is accepted that its ion, Ac 3+ , is the most voluminous trivalent cation of the periodic table. However, the structural data available on Ac 3+ compounds are scarce and have mainly been collected in the 1940-1960's, when actinide chemistry was still in its infancy, and have not been put in perspective with the advances in the chemistry of other elements, making it difficult to accurately evaluate its actual size and coordination chemistry. Here, we review progress made on the chemistry of lanthanides and actinides and reevaluate the structural data published on Ac 3+ since the era of the Manhattan Project. The data are combined across different spectroscopic and characterization methods and presented in the context of periodic trends. When considering crystallographic data, solution chemistry results, and the nuclear properties of actinium isotopes, it appears that some structural parameters ascribed to the Ac 3+ ion may have been overestimated. This review can guide researchers interested in actinide sciences and those who are pursuing the development of actinium-based radiotherapies, from isotope production to clinical trials.

Batch and column studies of radium, actinium, thorium and protactinium on CL resin in nitric acid, hydrochloric acid and hydrofluoric acid

CL resin (TrisKem International) was characterized for separations of radium, actinium, thorium and protactinium. Batch studies are presented for radium, actinium and thorium in HCl and HNO3 as well as for radium, thorium and protactinium in HF. The uptake of thorium from dilute HNO3 (≤ 10 -4 M) and dilute HCl (≤ 10 -4 M) was found to be very high (D w > 100 in HNO 3 and > 1000 in HCl). Kinetics studies on thorium were performed in HCl and HNO 3 to determine suitably for column separations. Six column separation studies are presented including the separation of trace thorium from radium and actinium in dilute HCl and HNO 3 , the separation of radium from 1 mg 232 Th in dilute HCl, the separation of thorium and radium in the presence of HCl and excess Cl-, the separation of radium, protactinium and thorium by retention of protactinium and the separation of protactinium and thorium by retention of thorium.

Tumor targeted alpha particle therapy with an actinium-225 labelled antibody for carbonic anhydrase IX

Selective antibody targeted delivery of α particle emitting actinium-225 to tumors has significant therapeutic potential. This work highlights the design and synthesis of a new bifunctional macrocyclic diazacrown ether chelator, H 2 MacropaSqOEt, that can be conjugated to antibodies and forms stable complexes with actinium-225. The macrocyclic diazacrown ether chelator incorporates a linker comprised of a short polyethylene glycol fragment and a squaramide ester that allows selective reaction with lysine residues on antibodies to form stable vinylogous amide linkages. This new H 2 MacropaSqOEt chelator was used to modify a monoclonal antibody, girentuximab (hG250), that binds to carbonic anhydrase IX, an enzyme that is overexpressed on the surface of cancers such as clear cell renal cell carcinoma. This new antibody conjugate (H 2 MacropaSq-hG250) had an average chelator to antibody ratio of 4 : 1 and retained high affinity for carbonic anhydrase IX. H 2 MacropaSq-hG250 was radiolabeled quantitatively with [ 225 Ac]Ac III within one minute at room temperature with micromolar concentrations of antibody and the radioactive complex is stable in human serum for >7 days. Evaluation of [ 225 Ac]Ac(MacropaSq-hG250) in a mouse xenograft model, that overexpresses carbonic anhydrase IX, demonstrated a highly significant therapeutic response. It is likely that H 2 MacropaSqOEt could be used to modify other antibodies providing a readily adaptable platform for other actinium-225 based therapeutics.

Capturing an elusive but critical element: Natural protein enables actinium chemistry

Actinium-based therapies could revolutionize cancer medicine but remain tantalizing due to the difficulties in studying and limited knowledge of Ac chemistry. Current efforts focus on small synthetic chelators, limiting radioisotope complexation and purification efficiencies. Here, we demonstrate a straightforward strategy to purify medically relevant radiometals, actinium(III) and yttrium(III), and probe their chemistry, using the recently discovered protein, lanmodulin. The stoichiometry, solution behavior, and formation constant of the 228 Ac 3+ -lanmodulin complex and its 90 Y 3+ / nat Y 3+ / nat La 3+ analogs were experimentally determined, representing the first actinium-protein and strongest actinide(III)-protein complex (sub-picomolar Kd) to be characterized. Lanmodulin’s unparalleled properties enable the facile purification recovery of radiometals, even in the presence of >10 +10 equivalents of competing ions and at ultratrace levels: down to 2 femtograms 90 Y 3+ and 40 attograms 228 Ac 3+ . The lanmodulin-based approach charts a new course to study elusive isotopes and develop versatile chelating platforms for medical radiometals, both for high-value separations and potential in vivo applications.

Cation exchange separation of radium and actinium using lactic acid DTPA buffer

The separation of actinium ( 228 Ac) from radium ( 223,228 Ra) on cation exchange resin with a diethylenetriaminpentaacetic acid-lactate buffer solution is demonstrated with series of columns. High yield, high radiopurity (~ 100%) separations of Ac from Ra are feasible with small columns in biologically compatible conditions and pHs. As Ac is eluted before Ra on these columns, further studies were performed to determine if this separation system could be applied to Ra/Ac isotope generators, but these were not successful. In conclusion, the separations presented in this work may be relevant for radiopharmaceutical purifications of 225 Ac which is typically obtained from its 225 Ra parent isotope.

Separation of radium and actinium using zirconia

Herein, zirconia was investigated as a radiation-resistant support for radium and actinium aqueous ion separations. Acetate and total metal ion concentrations were experimentally evaluated as factors that could affect retention using La and Ba surrogates. Pseudo-second order rate constants were derived and the uptake of La was determined to be endothermic. Elution profiles containing Ba, La, 228 Ra, 228 Ac, and 212 Pb are presented and discussed.

H 2 BZmacropa-NCS: A Bifunctional Chelator for Actinium-225 Targeted Alpha Therapy

Actinium-225 ( 225 Ac) is one of the most promising radionuclides for targeted alpha therapy (TAT). With a half-life of 9.92 days and a decay chain that emits four high-energy α particles, 225 Ac is well-suited for TAT when conjugated to macromolecular targeting vectors that exhibit extended in vivo circulation times. The implementation of 225 Ac in these targeted constructs, however, requires a suitable chelator that can bind and retain this radionuclide in vivo. Previous work has demonstrated the suitability of a diaza-18-crown-6 macrocyclic chelator H 2 macropa for this application. Building upon these prior efforts, in this study, two rigid variants of H 2 macropa, which contain either one (H 2 BZmacropa) or two (H 2 BZ 2 macropa) benzene rings within the macrocyclic core, were synthesized and investigated for their potential use for 225 Ac TAT. The coordination chemistry of these ligands with La 3+ , used as a nonradioactive model for Ac 3+ , was carried out. Both NMR spectroscopic and X-ray crystallographic studies of the La 3+ complexes of these ligands revealed similar structural features to those found for the related complex of H 2 macropa. Thermodynamic stability constants of the La 3+ complexes, however, were found to be 1 and 2 orders of magnitude lower than those of H 2 macropa for H 2 BZmacropa and H 2 BZ 2 macropa, respectively. The decrease in thermodynamic stability was rationalized via the use of density functional theory calculations. 225 Ac radiolabeling and serum stability studies with H 2 BZmacropa showed that this chelator compares favorably with H 2 macropa. Based on these promising results, a bifunctional version of this chelator, H 2 BZmacropa-NCS, was synthesized and conjugated to the antibody codrituzumab (GC33), which targets the liver cancer biomarker glypican-3 (GPC3). The resulting GC33-BZmacropa conjugate and an analogous GC33-macropa conjugate were evaluated for their 225 Ac radiolabeling efficiencies, antigen-binding affinities, and in vivo biodistribution in HepG2 liver cancer tumor-bearing mice. Although both conjugates were comparably effective in their radiolabeling efficiencies, [ 225 Ac]Ac-GC33-BZmacropa showed slightly poorer serum stability and biodistribution than [ 225 Ac]Ac-GC33-macropa. Altogether, these results establish H2BZmacropa-NCS as a new bifunctional chelator for the preparation of 225 Ac radiopharmaceuticals.

Actinium chelation and crystallization in a macromolecular scaffold

Abstract Targeted alpha therapy (TAT) pairs the specificity of antigen targeting with the lethality of alpha particles to eradicate cancerous cells. Actinium-225 [ 225 Ac; t 1/2 = 9.920(3) days] is an alpha-emitting radioisotope driving the next generation of TAT radiopharmaceuticals. Despite promising clinical results, a fundamental understanding of Ac coordination chemistry lags behind the rest of the Periodic Table due to its limited availability, lack of stable isotopes, and inadequate systems poised to probe the chemical behavior of this radionuclide. In this work, we demonstrate a platform that combines an 8-coordinate synthetic ligand and a mammalian protein to characterize the solution and solid-state behavior of the longest-lived Ac isotope, 227 Ac [t 1/2 = 21.772(3) years]. We expect these results to direct renewed efforts for 225 Ac-TAT development, aid in understanding Ac coordination behavior relative to other +3 lanthanides and actinides, and more broadly inform this element’s position on the Periodic Table.

Synergistic extraction of rare earth elements, actinium, americium, and barium with TTA and Pb resin from acetate buffer solutions

The uptake behavior of 88 Y, 140 Ba, 140 La, 141 Ce, 152 Eu, 228 Ac, and 241 Am was studied with batch uptake, kinetics and column studies in a synergistic extraction system with Pb resin and 2-thenoyltrifuoroacetone in acetate-ethanol solutions. The k' values are very high (>10,000) at near neutral pH (~5 to 6) for all of the radioisotopes studied. The kinetics are reasonably fast for column separations, though the uptake rate is dependent on the concentration of 2-thenoyltrifuoroacetone. Column studies were performed to demonstrate the separation of the trivalent actinides and lanthanides from barium and common fssion products ( 99 Mo, 103 Ru).

Actinium–DOTA coordination in water from hybrid ML/MM: Structure, free energies, and water-exchange pathways

Quantitative simulation of trivalent ƒ-block chelates in water remains challenging because bonded and non-bonded force-field models make different approximations for coordination structure, exchange dynamics, and ion–ligand interactions in highly charged systems. Here, we develop a hybrid machine-learning/molecular-mechanics (ML/MM) framework for Ac 3+ –DOTA in explicit solvent by training an E(3)-equivariant neural network potential (MACELES) on mechanically embedded QM/MM data for Ac aquo and Ac–DOTA species and coupling it to NAMD 2.14 with particle-mesh Ewald electrostatics. Nanosecond ML/MM trajectories remain numerically stable and preserve chelate integrity, yielding a compact DOTA inner shell with an inner-sphere water coordination number of CN Ac,O w ≈ 1.7 arising from a dynamic equilibrium between one- and two-water states (37.5% and 59.9% of frames; three waters 2.5%). A 5 ns potential of mean force shows two low-lying basins at CN Ac,O w ≈ 1 and CN Ac,O w ≈ 2. DFT end-state free energies are consistent with the ML/MM profile, and DFT minimum-energy paths provide a qualitative electronic-structure reference for the observed basin connectivity. State-resolved kinetics reveal picosecond water-exchange pathways that couple hydration changes to transient DOTA arm fluctuations, and training-set comparisons show that temperature-matched Ac–DOTA data optimize energy/force accuracy while more diverse solvated data improve charge prediction. Overall, the present hybrid ML/MM model provides a practical description of Ac 3+ –DOTA hydration thermodynamics and short-time exchange behavior in explicit water at MD-like cost.

Separation of radium and actinium in acetic acid/acetate solutions using TK101 resin

Here, the uptake and elution behavior of Ra and Ac was studied on TK101 resin using acetic acid and acetate buffer (acetic acid/lithium acetate) solutions. There is high extraction of Ra (>10 3 ) over a wide range of acetic acid concentrations (0.02 to 4 M) with slightly lower extraction (>10 2 ) in the acetate buffer solutions (pH 3.9 to 5.6). The Ac extraction is considerably lower with a maximum D w of ∼100, and negligible extraction at high acetic acid concentrations (> 1 M) and in the acetate buffer solutions. The difference in D w can be leveraged for highly efficient separations of Ac from Ra, with no detectable Ra in the Ac fractions and high Ac yields (∼100%). These separations may be useful for radiopharmaceutical applications as the Ac purity is extremely high and the elution conditions can be optimized to be biologically safe and appropriate for chelation to molecules relevant to targeted alpha therapy.

Metal Oxide Sorbents for the Separation of Radium and Actinium

The inorganic, normal-phase sorbents alumina, titania, and zirconia were evaluated and screened for the separation of Ba, La, 228 Ra, 228 Ac, and 212 Pb in a dilute acid matrix. The results show that the sorbents are capable of fractionating the ions between pH 1 and 8. A stationary zirconia column was used to isolate Ba from La and Ra from Ac/Pb; elution profiles of each separation are presented. The utility and implications of these sorbents are discussed with respect to large-scale production of 225 Ac from radium targets.
