CRISPR/Cas9-Assisted Transformation-Efficient Reaction (CRATER) for Near-Perfect Selective Transformation

The CRISPR (Clustered, Regularly Interspaced, Short Palindromic Repeats)/Cas9 system has revolutionized genome editing by providing unprecedented DNA-targeting specificity. Here we demonstrate that this system can be also applied in vitro to fundamental cloning steps to facilitate efficient plasmid selection for transformation and selective gene insertion into plasmid vectors by cleaving unwanted plasmid byproducts with a single-guide RNA (sgRNA)-Cas9 nuclease complex. Using fluorescent and chromogenic proteins as reporters, we demonstrate that CRISPR/Cas9 cleavage excludes multiple plasmids as well as unwanted ligation byproducts resulting in an unprecedented increase in the transformation success rate from approximately 20% to nearly 100%. Thus, this CRISPR/Cas9-Assisted Transformation-Efficient Reaction (CRATER) protocol is a novel, inexpensive, and convenient application to conventional molecular cloning to achieve near-perfect selective transformation.

On the variational equations for Householder transformations in feature selection

Results that suggest the possibility of using a sequential monotone process for solving the feature selection problem using Householder transformations are applied to the divergence separability criterion and an expression for the gradient of the divergence with respect to the generator of a single Householder transformation will be developed. This expression for the gradient is used in any number of differential correction schemes (iterators) that attempt to extremize the divergence. Data sets provided by the Earth Observations Division-JSC are used to demonstrate selecting the Householder transformations that generate the kxn matrix defining the best (in the sense of extremizing the divergence) k linear combinations of features. The tests allow initial comparisons to be made with results. In particular, this technique does not appear to require initial guesses for the iterator to be generated without replacement, exhaustive search, or other similar schemes.

Battery system and method for sensing and balancing the charge state of battery cells

A battery system utilizes a plurality of transformers interconnected with the battery cells. The transformers each have at least one transformer core operable for magnetization in at least a first magnetic state with a magnetic flux in a first direction and a second magnetic state with a magnetic flux in a second direction. The transformer cores retain the first magnetic state and the second magnetic state without current flow through said plurality of transformers. Circuitry is utilized for switching a selected transformer core between the first and second magnetic states to sense voltage and/or balance particular cells or particular banks of cells.

Spacecraft transformer and inductor design

The conversion process in spacecraft power electronics requires the use of magnetic components which frequently are the heaviest and bulkiest items in the conversion circuit. This handbook pertains to magnetic material selection, transformer and inductor design tradeoffs, transformer design, iron core dc inductor design, toroidal power core inductor design, window utilization factors, regulation, and temperature rise. Relationships are given which simplify and standardize the design of transformers and the analysis of the circuits in which they are used. The interactions of the various design parameters are also presented in simplified form so that tradeoffs and optimizations may easily be made.

Product Distribution Theory and Semi-Coordinate Transformations

Product Distribution (PD) theory is a new framework for doing distributed adaptive control of a multiagent system (MAS). We introduce the technique of "coordinate transformations" in PD theory gradient descent. These transformations selectively couple a few agents with each other into "meta-agents". Intuitively, this can be viewed as a generalization of forming binding contracts between those agents. Doing this sacrifices a bit of the distributed nature of the MAS, in that there must now be communication from multiple agents in determining what joint-move is finally implemented However, as we demonstrate in computer experiments, these transformations improve the performance of the MAS.

Landsat features for agricultural applications

This paper presents relationships among Landsat MSS bands and selected transformations of them, with emphasis on the Tasseled-Cap Transformation and its Brightness, Greenness variables. It also discusses relationships between reflectance measurements made in the Landsat spectral bands and actual Landsat data. Agronomically oriented analyses of reflectance measurements of wheat throughout a growing season are presented, with a comparison of various green measures, correlation with crop development stage, and examination of the effects of moisture stress. The final example addresses the use of transformed variables in a newly developed approach to forestry change detection.

Selective cytotoxicity of transformed cells but not normal cells by a sialoglycopeptide growth regulator in the presence of tumor necrosis factor

The tumor necrosis factor-alpha (TNF)-resistant, SV40-transformed, murine fibroblast cell lines, F5b and F5m, became sensitive to TNF-mediated cytolysis after treatment with a biologically active 18 kDa peptide fragment (SGP) derived from a 66-kDa parental cell surface sialoglycoprotein. Neither TNF nor the SGP alone exhibited cytotoxicity to the two SV40-transformed cell lines. However, Balb/c 3T3 cells, incubated with SGP alone or with SGP and TNF, were not killed. Therefore, SGP can selectively sensitize cells for TNF alpha-mediated cytotoxicity. This selective sensitization may be due to the previously documented ability of the SGP to selectively mediate cell cycle arrest.

Feature Extraction and Selection Strategies for Automated Target Recognition

Several feature extraction and selection methods for an existing automatic target recognition (ATR) system using JPLs Grayscale Optical Correlator (GOC) and Optimal Trade-Off Maximum Average Correlation Height (OT-MACH) filter were tested using MATLAB. The ATR system is composed of three stages: a cursory region of-interest (ROI) search using the GOC and OT-MACH filter, a feature extraction and selection stage, and a final classification stage. Feature extraction and selection concerns transforming potential target data into more useful forms as well as selecting important subsets of that data which may aide in detection and classification. The strategies tested were built around two popular extraction methods: Principal Component Analysis (PCA) and Independent Component Analysis (ICA). Performance was measured based on the classification accuracy and free-response receiver operating characteristic (FROC) output of a support vector machine(SVM) and a neural net (NN) classifier.

Universal and Automated Monte Carlo Method Code for Uncertainty Propagation in Metrology Databases

A method of providing transformed target points for integrating a component into an assembly includes collecting a set of component target points, collecting a set of assembly target points, identifying target points common to the set of component target points and the set of assembly target points; performing a specified number of Monte Carlo transformations of selected ones of the common target points to yield a set of transformed target points and vectors and an associated uncertainty value for each transformed target point and vector, and using certain ones of the transformed target points for integrating the component into the assembly based on the associated uncertainty value for each of the transformed target points.

Assessment of Shape Memory Alloys - From Atoms To Actuators - Via In Situ Neutron Diffraction

As shape memory alloys (SMAs) become an established actuator technology, it is important to identify the fundamental mechanisms responsible for their performance by understanding microstructure performance relationships from processing to final form. Yet, microstructural examination of SMAs at stress and temperature is often a challenge since structural changes occur with stress and temperature and microstructures cannot be preserved through quenching or after stress removal, as would be the case for conventional materials. One solution to this dilemma is in situ neutron diffraction, which has been applied to the investigation of SMAs and has offered a unique approach to reveal the fundamental micromechanics and microstructural aspects of bulk SMAs in a non-destructive setting. Through this technique, it is possible to directly correlate the micromechanical responses (e.g., internal residual stresses, lattice strains), microstructural evolutions (e.g., texture, defects) and phase transformation properties (e.g., phase fractions, kinetics) to the macroscopic actuator behavior. In this work, in situ neutron diffraction was systematically employed to evaluate the deformation and transformation behavior of SMAs under typical actuator conditions. Austenite and martensite phases, yield behavior, variant selection and transformation temperatures were characterized for a polycrystalline NiTi (49.9 at. Ni). As the alloy transforms under thermomechanical loading, the measured textures and lattice plane-level variations were directly related to the cyclic actuation-strain characteristics and the dimensional instability (strain ratcheting) commonly observed in this alloy. The effect of training on the shape memory characteristics of the alloy and the development of two-way shape memory effect (TWSME) were also assessed. The final conversion from a material to a useful actuator, typically termed shape setting, was also investigated in situ during constrained heatingcooling and subsequent shape recovery experiments. Neutron diffraction techniques are also being applied to the investigation of novel high temperature SMAs with the objective of designing alloys with better stability, higher transition temperatures and ultimately superior durability.

Programmable remapper with single flow architecture

The invention relates to image processing systems and methods and in particular to a machine which accepts a real time video image in the form of a matrix of picture elements (pixels) and remaps such image according to a selectable one of a plurality of mapping functions to create an output matrix of pixels. Such mapping functions, or transformations, may be any one of a number of different transformations depending on the objective of the user of the system. The system remaps input images from one coordinate system to another using a set of look-up tables for the data necessary for the transform. The transforms, which are operator selectable, are precomputed and loaded into massive look-up tables. Input pixels, via the look-up tables of any particular transform selected, are mapped into output pixels with the radiance information of the input pixels being appropriately weighted. An earlier embodiment of the system included two parallel processors: a collective processor which mapped multiple input pixels into a single output pixel and an interpolative processor. The interpolative processor performed an interpolation among pixels in the input image where a given input pixel may affect the value of many output pixels. Several advantages are provided over previous embodiments in that the two distinct processors are replaced by a single processor capable of performing both types of operations (collective and interpolative) with no more complexity. Previously, there has existed no image processor or 'remapper' that can operate with sufficient speed and flexibility to permit investigating different transformation patterns in real time.

Karyotyping of Chromosomes in Human Bronchial Epithelial Cells Transformed by High Energy Fe Ions

Lung cancer induced from exposure to space radiation is believed to be one of the most significant health risks for long-term space travels. In a previous study, normal human bronchial epithelial cells (HBECs), immortalized through the expression of Cdk4 and hTERT, were exposed to gamma rays and high energy Fe ions for the selection of transformed clones induced by low- and high-LET radiation. In this research, we analyzed chromosome aberrations in these selected clones for genomic instability using the multi-color fluorescent in situ hybridization (mFISH), as well as the multi-banding in situ hybridization (mBAND) techniques. In most of the clones, we found chromosomal aberrations involving translocations between different chromosomes, with several of the breaks occurred in the q-arm of chromosome 3. We also identified copy number variations between the transformed clones and the parental HBEC cells regardless of the exposure condition. Our results indicated that the chromosomal aberrations in low- and high radiation-induced transformed clones are inadequately different from spontaneous soft agar growth. Further analysis is underway to reveal the genomic instability in more transformed clones

A graphical, rule based robotic interface system

The ability of a human to take control of a robotic system is essential in any use of robots in space in order to handle unforeseen changes in the robot's work environment or scheduled tasks. But in cases in which the work environment is known, a human controlling a robot's every move by remote control is both time consuming and frustrating. A system is needed in which the user can give the robotic system commands to perform tasks but need not tell the system how. To be useful, this system should be able to plan and perform the tasks faster than a telerobotic system. The interface between the user and the robot system must be natural and meaningful to the user. A high level user interface program under development at the University of Alabama, Huntsville, is described. A graphical interface is proposed in which the user selects objects to be manipulated by selecting representations of the object on projections of a 3-D model of the work environment. The user may move in the work environment by changing the viewpoint of the projections. The interface uses a rule based program to transform user selection of items on a graphics display of the robot's work environment into commands for the robot. The program first determines if the desired task is possible given the abilities of the robot and any constraints on the object. If the task is possible, the program determines what movements the robot needs to make to perform the task. The movements are transformed into commands for the robot. The information defining the robot, the work environment, and how objects may be moved is stored in a set of data bases accessible to the program and displayable to the user.

Circumstances for Pluto-Charon mutual events in 1988

Physical parameters are tabulated for 89 Pluto-Charon mutual events occurring during the 1988 opposition. A primary star and a check star have been selected as comparison stars for events occurring prior to the 1988 opposition. Standardization of the comparison star 1987 Primary has provided a B magnitude of 12.3093 + or - 0.0013 and a V magnitude of 11.4215 + or - 0.0013. The designations, positions, and preliminary magnitudes and colors are also given for two transformation stars selected in order to aid in determination of the color terms necessary to convert the instrumental magnitudes of observers to the standard system.
