Low-chromophore lignin isolation from natural biomass with polyol-based deep eutectic solvents

When attempting to obtain light-color lignin from lignocellulosic biomass or industrial lignin, the available options based on chemical or morphological modification suffer from low yield, high cost, and lack of availability at the required scales. In this study, we adopted a polyhydric-alcohol-based deep eutectic solvent (PA-DES) to directly extract light-color lignin from natural biomass, which is even whiter than native cellulolytic enzyme lignin (CEL). The isolated lignin possessed a high recovery yield (97.36%), regular micro-spherical morphology, enriched β-ether linkage of 58/100Ar, low phenolic hydroxyl content of 1.25 mmol g -1 , minimal carbonyl content of 0.70 mmol g -1 , and less condensed structures, thus yielding a lower content of chromophores. Further, this lignin showed excellent sunscreen effects, which could enhance the SPF of a commercial sunscreen from 15 to 40 with only 5 wt% addition. This study can provide essential guidance for the scale-up production of light-color lignin and obtaining near-complete digestible cellulose for further saccharification.

Efficient Fast Fractionation of Biomass Using a Diol-Based Deep Eutectic Solvent for Facilitating Enzymatic Hydrolysis and Obtaining High-Quality Lignin

Current DES pretreatment is often performed under relatively severe conditions with high temperature, long time, and high DES usage. This work studied a short-time diol DES (deep eutectic solvent) pretreatment under mild conditions to fractionate the bamboo, facilitate enzymatic hydrolysis, and obtain high-quality lignin. At an optimized condition of 130 °C for only 10 min, lignin and xylan removal reached 61.34% and 84.15 %, with residual glucan showing a ~90% enzymatic hydrolysis yield. Equally important, the dissolved lignin could be readily recovered with 97.51% yield, exhibiting 96.65 % β-O-4 preservation. The fractionation and lignin protection mechanisms were unveiled by XRD, FTIR, cellulose-DP, 2D HSQC NMR, 31 P NMR and GPC analysis. Finally, this study highlighted that short-time fractionation of bamboo can be achieved by a diol-based DES which is an ideal strategy to upgrade the lignocellulose biomass for high enzymatic hydrolysis yields and high-quality lignin stream.

A novel mineral-acid free biphasic deep eutectic solvent/γ-valerolactone system for furfural production and boosting the enzymatic hydrolysis of lignocellulosic biomass

The failure of hemicellulose valorization in a deep eutectic solvent (DES) pretreatment has become a bottleneck that challenges its further development. Here, to address this issue, this study developed a DES/GVL (γ-valerolactone) biphasic system for effective hemicellulose-furfural conversion, enhanced cellulose saccharification and lignin isolation. The results indicated that the biphasic system could significantly improve the lignin removal (as high as 89.1%), 86.0% higher than the monophasic DES, accompanied by ~100% hemicellulose degradation. Notably, the GVL in the biphasic solvent restricted the condensation of hemicellulose degradation products, which as a result generated large amount of furfural in the pretreatment liquid with a yield of 68.6%. With the removal of hemicellulose and lignin, cellulose enzymatic hydrolysis yield was boosted and reached near 100%. This study highlighted that the novel DES/GVL is capable of fractionating the biomass and benefiting their individual utilization, which could provide a new biorefinery configuration for a DES pretreatment.

Direct Extraction of Uniform Lignin Microspheres from Bamboo Using a Polyol-Based Deep Eutectic Solvent

Conventional preparation of lignin micro-/nano-particles (LMNPs) often needs multiple steps accompanied by high cost, low efficiency, and low yield. Herein, this study established a polyhydric alcohol-based deep eutectic solvent (PA-DES) fractionation for direct LMNP assembly from raw lignocellulose. The results indicated that lignin was significantly extracted by 41.55–70.33% depending on the PA-DES varieties. With lignin removal, the substrate showed high glucan enzymatic saccharification from 70.44 to 100%. Specially, over 90% lignin could be recovered in each PA-DES, and the obtained LMNPs showed quite uniform spherical morphology with a tunable size of 550–3254 nm and narrow polydispersity index (PDI) of 0.295–0.398. The proposed DES showed a near theoretical recovery yield, which still featured excellent fractionation performance even after three cycles of reuse. In conclusion, the findings highlight the simultaneous utilization of cellulose and lignin in a one-pot biomass fractionation under mild conditions with sugars and LMNPs as the coproducts.

One-pot generation of lignin microspheres and digestible substrate with a polyol-DES pretreatment in high solid loading

Previous lignin microspheres (LMS) preparation needs multiple steps with very low yield and high cost. Herein, we developed a high-solid deep eutectic solvent (DES) pretreatment for an effective lignin fractionation and enzymatic saccharification of moso bamboo under mild temperature (110 °C) with high-solid loading. Lignin was significantly removed from the plant cell wall, and cellulose properties (e.g., crystallinity and degree of polymerization) were also altered during the pretreatment. As a result, the enzymatic digestibility of the pretreated bamboo was dramatically increased. Uniform micro-spherical lignin was directly produced from the pretreatment system, and its particle size could be regulated by controlling the solid loadings and pretreatment temperatures. The lignin microspheres formation mechanism was investigated by analyzing the lignin's size distribution, molecular weight distribution, chemical structure, and hydrophobicity. The DES showed excellent recyclability, and the recycled DES could still remove 42.78% lignin even after 7th circulation associated with 100% glucan saccharification. Further, the mass balance based on 1000 g biomass showed that 196.28 g LMS was directly recovered, which exhibited a high RhB adsorption. Besides, 404.91 g glucose and 36.67 g xylose were obtained after the enzymatic saccharification process. Specifically, GAPI analysis exhibited a near total green and yellow portions of the pictogram, indicating our DES process was green enough to make this biorefinery sustainable. Overall, the proposed DES generated synergistic productions of digestible solid and LMS which could contribute to establish a green and sustainable biorefinery sequence with diverse outputs in one pot.

A high-solid DES pretreatment using never-dried biomass as the starting material: towards high-quality lignin fractionation

This study investigated a high-solid diol deep eutectic solvent (DES) pretreatment using a wet substrate as the starting material. This pretreatment led to a remarkable glucan saccharification of 94.8% with efficient lignin and xylan removal (as high as 63.1% and 73.0%, respectively). Here the chemical structures of the substrates were analyzed comprehensively to reveal the impact of the pretreatment. In addition, over 90% of the removed lignin was recovered from the pretreated liquid, which exhibited a well-preserved β-O-4 structure (46–56/100Ar). The protection mechanism of our DES was investigated by 2D HSQC NMR, GPC, and 31 P NMR analysis. This study emphasized that diol-based DES pretreatment of undried lignocellulosic biomass at a high-solid loading can significantly utilize both carbohydrates and lignin fractions with high saccharification yields and high-quality lignin as a co-product.

Alkali-facilitated deep eutectic solvent for effective bamboo saccharification

In this report, a Na 2 S promoted deep eutectic solvent (DES) was established to reduce the natural recalcitrance of moso bamboo (MB) and improve the subsequent enzymatic saccharification. It was found that the addition of Na 2 S (Choline chloride/Ethylene glycol/Na 2 S) dramatically promoted the deconstructions of lignin with highest removal of 74.67 %, but at the same time preserved glucan and hemicellulose to the maximum extent. With the fractionation, the enzymatic saccharification yield of pretreated MB can reach 100 % under the pretreatment condition of 140 °C, and lignin could be readily recovered with a high yield of 81.47 %. The proposed DES is superior to normal alkaline DES in terms of the higher lignin removal and recovery yield, carbohydrate preservation and enzymatic digestibility, which indicated Na 2 S as a novel and powerful reinforcer enhancing the DES fractionation efficiency.

Utilization of guaiacol-based deep eutectic solvent for achieving a sustainable biorefinery

This study proposed a renewable deep eutectic solvent (DES) pretreatment using lignin-derived guaiacol as the hydrogen bond donor. The DES showed excellent biomass fractionation efficiency after the incorporation of trace AlCl 3 as the reinforcer, which removed 79.1 % lignin while preserving more than 90 % glucan. The pretreated bamboo exhibited 96.2 % glucan enzymatic hydrolysis yield at only 110 °C. The physicochemical properties of the pretreated solids were comprehensively investigated to explain how the DES fractionation overcame the biomass recalcitrance. The regenerated lignin from the DES pretreatment was also analyzed, which revealed that lignin β-O-4 bond was significantly cleaved. In conclusion, this guaiacol-based DES could greatly contribute to establish a closed-loop biorefinery sequence with high lignin fractionation efficiency and great solvent recyclability.
