Small-molecule modulation of β-arrestins

β-Arrestins are multifunctional regulators of G-protein-coupled receptor (GPCR) signalling and orchestrate diverse downstream signalling events and physiological responses across the GPCR superfamily. Although GPCR pharmacology has advanced to target orthosteric and allosteric sites, as well as G proteins and GPCR kinases, direct chemical tools to modulate β-arrestin activities have remained conspicuously absent. Here we report the identification of small-molecule inhibitors that selectively target β-arrestins and delineate their mechanism of action through integrated pharmacological, biochemical, biophysical and structural analyses. These inhibitors disrupt β-arrestin engagement with agonist-activated GPCRs, impairing desensitization, internalization and β-arrestin-dependent physiological functions while sparing G protein–receptor coupling. Cryo-electron microscopy, molecular dynamics simulations and structure-guided mutagenesis reveal that one modulator, Cmpd-5, engages a pocket within the central crest of β-arrestin1 formed by the middle, C and lariat loops, a critical receptor-binding interface, stabilizing a distinct conformation that is incompatible with full β-arrestin–receptor engagement. Together, these findings establish a mechanistic framework for β-arrestin modulation, reveal a novel allosteric site for structure-based drug design, and open new avenues for transducer-targeted, pathway-specific GPCR therapeutic agents.

An Accreting Supermassive Black Hole Buried in a Faint Dwarf Galaxy

Abstract In the last decade, there have been several discoveries of active galactic nuclei (AGN) in dwarf galaxies including an AGN in an ultracompact dwarf galaxy with a black hole mass >10 6 M ⊙ . However, finding a supermassive black hole (SMBH) in a dwarf low surface brightness (LSB) galaxy is rare. We report the discovery of a Seyfert type 2 class AGN that is associated with a nuclear SMBH of mass ∼6.5 × 10 6 M ⊙ in a dwarf LSB galaxy (μ 0,r > 23.8 mag arcsec −2 ) that we denote by MJ0818+2257. The galaxy was previously thought to be an outlying emission blob around the large spiral galaxy LEDA 1678924. In our current analysis, which includes the detection of the optical counterpart of MJ0818+2257, we study its ionized gas kinematics and find that the dynamical mass within the ionized gas disk is ∼5.3 × 10 9 M ⊙ . This is comparable to its stellar mass, which is ∼3 × 10 9 M ⊙ , and suggests that MJ0818+2257 is moderately dark matter dominated within the stellar disk. The SMBH-mass-to-galaxy-stellar-mass ratio isM BH /M(*) > 0.022, which is high compared to disk galaxies. Our detection of an SMBH in a bulgeless LSB dwarf galaxy raises questions about the growth of SMBHs in low-luminosity galaxies and suggests the possibility of detecting heavy seed black holes from early epochs in LSB dwarf galaxies in the low-redshift Universe.

Identification of potent inhibitors of JUN N-terminal kinases for treatment of endometriosis and associated pain

Endometriosis, defined as the ectopic growth of endometrial tissue outside of the uterine cavity, is an inflammatory and hormone-dependent disease that causes excruciating pelvic pain, infertility, and significantly decreases quality of life in affected patients. The JUN N-terminal kinases (JNKs) are a leading class of nonhormonal therapeutic targets that have been validated in preclinical models of endometriosis and in a Phase 1/2 clinical trial. Despite their therapeutic potential, JNK inhibitors with increased potency and specificity are needed to address the inflammatory pathology of endometriosis and to prevent disease progression. Leveraging a DNA-encoded chemical library collection of ~4 billion compounds, we identified lead inhibitor CDD-2428 and optimized derivatives, CDD-2728 and CDD-3013, with excellent binding affinity to JNK1-3 (K d = 0.12 to 3.7 nM), enhanced selectivity, metabolic stability, and cellular permeability. Crystallographic and biochemical studies confirmed that CDD-3013 exhibited superior kinase selectivity with improved efficacy compared to existing JNK inhibitors. In primary endometriosis cell models, CDD-2728 and CDD-3013 suppressed JNK-dependent inflammatory signaling, dampening pathways linked to pain, invasion, angiogenesis, and macrophage recruitment. In an endometriosis mouse model, both CDD-2728 and CDD-3013 reduced endometriotic lesion size, macrophage infiltration, and cellular proliferation, showing in vivo efficacy. When tested in a lipopolysaccharide-induced hyperalgesia model, CDD-2728 and CDD-3013 decreased markers of induced pain, as measured by changes in a dynamic weight bearing test and Grimace scores. These findings nominate CDD-2728 and CDD-3013 as potent, nonhormonal therapeutic candidates for endometriosis with broad anti-inflammatory and analgesic activity, addressing a critical unmet clinical need.

Adventures in Research: A History of Ames Research Center, 1940-1965

The present document, which I have called a history-though it certainly was not written for historians-was prepared for and at the request of the NASA Ames Research Center which, in sponsoring the project, acted with the encouragement of NASA Headquarters. It is an account of the establishment, evolutionary development, and activities of the Ames Research Center covering a period characterized by unprecedented scientific and technological revolution. The writing of this book about the Ames Research Center was undertaken with considerable pleasure by one who for many years was employed by the Center’s parent organization NASA and, before that, NACA; who had closely followed the Center’s growth from the beginning; and who was personally acquainted with, and who held in some affection, the members of its founding staff. These facts are given to alert readers to the presence of bias and the inaccuracies stemming therefrom. It should be emphasized that this work is a history only of the Ames Research Center. It is not a history of the U.S. Government, of NACA or NASA, of other research centers, of the military services, of industry, or of world events. Information on these peripheral subjects is introduced merely to provide the reader with a generalized feeling for the local and world environment in which the formation and subsequent activities of the Ames Research Center took place. The background material provided is notably incomplete and in some instances reveals the bias of the NACA/NASA community at the time the events took place. Frequently, also, it reflects the personalized viewpoint of the author who was a close observer of many of these events.

FAST Discovery of a Fast Neutral Hydrogen Outflow

Abstract In this letter, we report the discovery of a fast neutral hydrogen outflow in SDSS J145239.38+062738.0, a merging radio galaxy containing an optical type I active galactic nucleus (AGN). This discovery was made through observations conducted by the Five-hundred-meter Aperture Spherical radio Telescope (FAST) using redshifted 21 cm absorption. The outflow exhibits a blueshifted velocity likely up to ∼−1000 km s −1 with respect to the systemic velocity of the host galaxy with an absorption strength of ∼−0.6 mJy beam −1 corresponding to an optical depth of 0.002 atv= −500 km s −1 . The mass outflow rate ranges between 2.8 × 10 −2 and 3.6M ⊙ yr −1 , implying an energy outflow rate ranging between 4.2 × 10 39 and 9.7 × 10 40 erg s −1 , assuming 100 K s< 1000 K. Plausible drivers of the outflow include the starbursts, AGN radiation, and radio jet, the last of which is considered the most likely culprit according to the kinematics. By analyzing the properties of the outflow, AGN, and jet, we find that if the Hioutflow is driven by the AGN radiation, the AGN radiation does not seem powerful enough to provide negative feedback, whereas the radio jet shows the potential to provide negative feedback. Our observations contribute another example of a fast outflow detected in neutral hydrogen and demonstrate the capability of FAST in detecting such outflows.

Analysis of the Erosion of Cubic Centered and Face Centered Metallic Materials [Analyse de L'erosion de Matereaux Metalliques de Structures Cubiques Centree et Face Centree]

A generalized law of erosion, in nondimensional form, is proposed. The aforementioned explicitly gives an account of the nature and the microstructure of the material impacted, the angle of impact of the particles, and their hardness. The erosion behavior of various steels, pig iron, and cast iron is examined as well as the behavior of new ordered intermetallic materials, Fe-25% Al-(Zr, Cr, B). The concern of such alloys in an erosion-corrosion environment is shown. --- Une loi généralisée d'usure par érosion, sous forme non dimensionnelle, est proposée. Celle-ci rend compte explicitement de la nature et de la microstructure du matériau impacté, de l'angle d'impact des particules, de leur acuité at de leur durete. Le comportement à l'érosion de divers aciers et fontes est examiné ainsi que la tenue de nouveaux matériaux intermétalliques ordonnés Fe-25% Al - (Zr, Cr, B). Aerospace Battery Activities at NASA/Goddard Space Flight Center

Goddard Space Flight Center has "pioneered" rechargeable secondary battery design, test, infusion and in-orbit battery management among NASA installations. Nickel cadmium batteries of various designs and sizes have been infused for LEO, GEO and Libration Point spacecraft. Nickel-Hydrogen batteries have currently been baselined for the majority of our missions. Li-Ion batteries from ABSL, JSB, SaFT and Lithion have been designed and tested for aerospace application.

Core-Centering of Compound Drops in Capillary Oscillations: Observations on USML-1 Experiments in Space

Using the existing inviscid theories, an attempt is made to explain the centering of the oscillating liquid shell. Experiments on liquid shells and liquid-core compound drops were conducted using acoustic levitation, in a low-gravity environment during a Space Shuttle flight. It was observed that their inner and outer interfaces became concentric when excited into capillary oscillations. Using the existing inviscid theories, and attempt is made to explain the centering of the oscillating liquid shell. It is concluded that viscosity needs to be considered in order to provide a realistic description of the centering process.

This document contains the proceedings of the 21st annual Battery Workshop held at Goddard Space Flight Center, Greenbelt, Maryland on November 1-3, 1988. The Workshop attendees included manufacturers, users, and government representatives interested in the latest developments in battery technology as they relate to high reliability operations and aerospace use. The subjects covered included battery testing methodologies and criteria, life testing of nickel-cadmium cells, testing and operation of nickel-hydrogen batteries in low earth orbit, and nickel-hydrogen technology issues and concerns.
