Ongoing Cooperative Engagement Facilitates Agile Pandemic and Outbreak Response: Lessons Learned Through Cooperative Engagement Between Uganda and the United States
Pathogens threaten human lives and disrupt economies around the world. This has been clearly illustrated by the current COVID-19 pandemic and outbreaks in livestock and food crops. Here, to manage pathogen emergence and spread, cooperative engagement programs develop and strengthen biosafety, biosecurity, and biosurveillance capabilities among local researchers to detect pathogens. In this case study, we describe the efforts of a collaboration between the Los Alamos National Laboratory and the Uganda Virus Research Institute, the primary viral diagnostic laboratory in Uganda, to implement and ensure the sustainability of sequencing for biosurveillance. We describe the process of establishing this capability along with the lessons learned from both sides of the partnership to inform future cooperative engagement efforts in low- and middle-income countries. We found that by strengthening sequencing capabilities at the Uganda Virus Research Institute before the COVID-19 pandemic, the institute was able to successfully sequence SARS-CoV-2 samples and provide data to the scientific community. We highlight the need to strengthen and sustain capabilities through in-country training, collaborative research projects, and trust.

Getting my team members to all agree that we needed to pursue possible solutions for our documentation review process was an idea that came across very well with the team. This specific topic has be something that as a team we have been having struggles with recently due to the shear amount of documents that have been flowing through the team’s hands to review. We all realized that the process had areas that we could change that would eliminate confusion for new employees, reduce backload, and improve efficiency and quality of the documentation reviews. The key personnel on our team that have the most direct control and impact over this process are the production control specialists, documentation preparation and review is their primary job function. They are denoted in the action pack as “doc spec 1” and this individual has 12 years of expertise in their role. The engineers are denoted as “engineer 1, 2, 3” respectively. Engineer 1 has 10 years of experience in this team, engineer 3 has 38 years of experience on the team, and engineer 2 is myself with 5 years of expertise with this team and 5 with the department of defense.

Energy Northwest — Advanced Grid Interactive Load Efficiency (AGILE) : A Techno-economic Assessment
The Advanced Grid Interactive Load Efficiency project team, led by Energy Northwest, was awarded funding as part of the fourth round of the Clean Energy Fund program to investigate, co-create, and complete a preliminary design for grid-interactive efficient buildings (GEBs) for a number of schools served by Grays Harbor PUD. The project team has evaluated different combinations of systems at selected school buildings and the potential to respond to grid and customer needs, and develop a preliminary system technical design, including specifying desired GEB technology options, determining communication and electrical interface requirements, assessing control software sensor requirements, and interfacing with building management systems. PNNL led the techno-economic assessment efforts, leveraging its advanced modeling and analytical methods and tools developed for building-to-grid integration. This report documents the economic assessment for different GEB design options, encompassing an in-depth examination of use cases and value propositions, assumptions and inputs, models, methods, case studies, as well as key findings.
Carbon optimized production of 3-hydroxypropionic acid in the Agile BioFoundry organism Rhodosporidium toruloides
The modern global economy relies heavily on carbon-based products that are derived from petroleum, which presents sustainability, resource management, and greenhouse gas exacerbated climate change challenges. Due to these challenges, there is the need for a global industrial transition towards green and sustainable production. Microbial production of valuable chemicals from renewable biomass represents one promising route. However, high-volume low-value products such as commodity chemicals are still difficult to make profitable. One fundamental bottleneck is a waste of more than 1/3 of the feedstock carbon as CO 2 in the fermentation process. Here the project focuses on fundamentally reconfiguring the metabolism to reduce CO 2 loss in central metabolic pathways thereby also improving bioproduct yields. Here we present technologies to prevent CO 2 loss and balance reducing equivalents within the cell to enable complete conversion of glucose from renewable feedstocks into bioproducts.
