Droplet microfluidics for synthetic biology

Here, synthetic biology is an interdisciplinary field that aims to engineer biological systems for useful purposes. Organism engineering often requires the optimization of individual genes and/or entire biological pathways (consisting of multiple genes). Advances in DNA sequencing and synthesis have recently begun to enable the possibility of evaluating thousands of gene variants and hundreds of thousands of gene combinations. However, such large-scale optimization experiments remain cost-prohibitive to researchers following traditional molecular biology practices, which are frequently labor-intensive and suffer from poor reproducibility. Liquid handling robotics may reduce labor and improve reproducibility, but are themselves expensive and thus inaccessible to most researchers. Microfluidic platforms offer a lower entry price point alternative to robotics, and maintain high throughput and reproducibility while further reducing operating costs through diminished reagent volume requirements. Droplet microfluidics have shown exceptional promise for synthetic biology experiments, including DNA assembly, transformation/transfection, culturing, cell sorting, phenotypic assays, artificial cells and genetic circuits.

Evaluation of Optimal Net Load Management in Microgrids Using Hardware-in-the-Loop Simulation

This paper presents the performance evaluation of a net load management (NLM) engine that balances load and generation in an isolated community to power a critical facility after a grid interruption event (e.g., the loss of a large generation unit). This NLM engine is particularly important for microgrid systems because it provides a high-speed, cost-optimal control solution to coordinate grid-forming inverters and to dispatch grid-following inverters and deferrable loads in microgrid systems to enhance grid resilience and reliability. The NLM algorithm cost-optimally dispatches the grid-following inverters and deferrable loads based on the demanded power and load priorities, and the grid-forming inverters use droop control to form system voltages and share active and reactive power. A controller-hardware-in-the-loop platform is developed to evaluate the control performance of the NLM algorithm with two sequential contingency events of lost generation units. The experimental results indicate that the NLM engine can maintain system stability, achieve the targeted system voltage and frequency, and balance load and generation to serve the critical facility with improved system resilience and reliability.

# 2023 University of Puerto Rico at Mayagüez National Institute for Congestion Reduction Study The 2023 University of Puerto Rico at Mayagüez National Institute for Congestion Reduction (NCIR) Study provided insight into the travel patterns and associated energy consumption of participants. Study results helped researchers identify opportunities for the development of policies that could incentivize the use of alternative modes of travel such as transit and micromobility. Such travel modes reduce congestion by reducing the miles traveled by privately owned vehicles in urban and rural areas. The [National Institute for Congestion Reduction](https://nicr.usf.edu/) provides multimodal congestion reduction strategies through real-world deployments that leverage advances in technology, big data science, and innovative transportation options to optimize the efficiency and reliability of the transportation system for all users. ## Data Collection Agency The University of Puerto Rico at Mayagüez conducted the study. ## Survey Methodology The study was conducted in Spanish. Data collection was enabled via the open-source [NREL OpenPATH platform](https://www.nrel.gov/transportation/openpath). The resulting dataset consists of partially automated travel diaries—combining sensed and surveyed data reflecting patterns of multimodal, end-to-end, individual human mobility—as well as demographic and socioeconomic information from the 17 participants. ## Survey Records, Data, and Documentation Study records include 17 participants. The total number of trips was 458 and total non-air-miles traveled was approximately 1,469.

The dynamics and control of large flexible space structures, 3. Part A: Shape and orientation control of a platform in orbit using point actuators

The dynamics, attitude, and shape control of a large thin flexible square platform in orbit are studied. Attitude and shape control are assumed to result from actuators placed perpendicular to the main surface and one edge and their effect on the rigid body and elastic modes is modelled to first order. The equations of motion are linearized about three different nominal orientations: (1) the platform following the local vertical with its major surface perpendicular to the orbital plane; (2) the platform following the local horizontal with its major surface normal to the local vertical; and (3) the platform following the local vertical with its major surface perpendicular to the orbit normal. The stability of the uncontrolled system is investigated analytically. Once controllability is established for a set of actuator locations, control law development is based on decoupling, pole placement, and linear optimal control theory. Frequencies and elastic modal shape functions are obtained using a finite element computer algorithm, two different approximate analytical methods, and the results of the three methods compared.

eVTOL Vehicle-Agnostic Instrument Flight Procedures Test Plan

NASA Advanced Air Mobility National Campaign is researching the utility of electric vertical takeoff and land (eVTOL) advanced air mobility (AAM) instrument flight procedures. The result will be dynamic and tailored procedures that align to the following modus operandi: maximize safety, optimize efficiency, support passenger comfort and minimize acoustics. This is achieved through dynamic airspace procedure design, which is a modular approach to create an airspace construct that customizes procedures to vehicle design and configuration, operation and environmental conditions. The test plan supports different eVTOL platforms and envisioned operations for flight test or simulation and may be leveraged by AAM aircraft manufacturers and operators for any given aircraft, location and operation.

Stripping off the implementation complexity of physics-based model predictive control for buildings via deep learning

Over the past decade, model predictive control (MPC) has been considered as the most promising solution for intelligent building operation. Despite extensive effort, transfer of this technology into practice is hampered by the need to obtain an accurate controller model with minimum effort, the need of expert knowledge to set it up, and the need of increased computational power and dedicated software to run it. A promising direction that tackles the last two problems was proposed by approximate explicit MPC where the optimal control policies are learned from MPC data via a suitable function approximator, e.g., a deep learning (DL) model. The main advantage of the proposed approach stems from simple evaluation at execution time leading to low computational footprints and easy deployment on embedded HW platforms. We present the energy savings potential of physics-based (also called 'white-box') MPC applied to an office building in Belgium. Moreover, we demonstrate how deep learning approximators can be used to cut the implementation and maintenance costs of MPC deployment without compromising performance. We also critically assess the presented approach by pointing out the major challenges and remaining open-research questions.

Evaluation of Optimal Net Load Management in Microgrids Using Hardware-in-the-Loop Simulation: Preprint

This paper presents the performance evaluation of a net load management (NLM) engine that balances load and generation in an isolated community to power a critical facility after a grid interruption event (e.g., lost a large generation unit). The NLM engine is a central dispatch control system that provides high-speed cost-optimal coordination of a set of net loads (combination of generation and deferrable loads) connected to the same electrical network. The NLM algorithm cost-optimally dispatches the grid-following inverters and deferrable loads based on the demanded power and load priorities, and the grid-forming inverters use droop control to form system voltages and share active and reactive power. A controller-hardware-in-the-loop platform is developed to evaluate the control performance of the NLM algorithm. The experimental results indicate that the NLM engine can achieve the targeted system voltage and frequency and balance load and generation to serve the critical facility.

Evaluation of Optimal Net Load Management in Microgrids Using Hardware-in-the-Loop Simulation

This presentation discusses the performance evaluation of a net load management (NLM) engine that balances load and generation in an isolated community to power a critical facility after a grid interruption event (e.g., the loss of a large generation unit).
