Creating a Training Dataset for Semantic Segmentation of Canal Networks for Irrigation Modernization

Canal infrastructure has provided critical irrigation water to the western United States for over a century. To continue providing vital water resources to the semi-arid West, irrigation systems must undergo maintenance and modernization. Many canal companies are resource-constrained, and because funding opportunities often require detailed knowledge of existing infrastructure, they can struggle to secure financial capital. We address this problem by creating training data for a semantic segmentation deep learning model to map canal networks throughout the western United States. To create a diverse and robust training dataset, we labelled 1-m NAIP imagery with the locations of no canals, wet canals, and dry/vegetated canals. Since creating these datasets is time consuming, we first developed a preprocessing methodology to identify canals within our four study areas. We used NAIP imagery and provided canal centerline data to buffer, standardize, and cluster the imagery, automating the labeling process as much as possible. However, this still required manual cleaning and manual classification of canal type. Challenges arose when canals were interrupted (e.g., road culverts or piped sections) or when nearby features shared similar characteristics (e.g., irrigated fields, trees, and shadows). Combining automated preprocessing with manual refinement produced four detailed canal masks to be used in the semantic segmentation model developed by Richard Tapia.

Investigation of competitive sorption and plasticization of hyperaged CANAL ladder polymers for acid gas purification

Identifying membrane materials that have exceptional separation performance and stability to complex CO 2 -containing mixtures is a pressing topic in separation science. In this work, the membrane separation performance for a recently discovered class of contorted polymers synthesized via catalytic arene-norbornene annulation (CANAL) polymerization is presented. These CANAL polymers achieve high CO 2 /CH 4 selectivity of 68 after physical aging (up to ∼1 year), which significantly augments the size-sieving capabilities of the membranes. Binary CO 2 /CH 4 and ternary H 2 S/CO 2 /CH 4 testing result in a 41 % and 50 % enhancement in selectivities, respectively, for hyperaged (∼1 year) contorted CANAL polymers, highlighting their size-sieving capabilities. The remarkably high CO 2 /CH 4 mixed-gas and combined acid gas (CAG, (CO 2 +H 2 S)/CH 4 ) selectivities of 88 and 95, respectively, for CANAL-Me-S 5 F in particular surpass both the 2018 CO 2 /CH 4 mixed-gas and CAG upper bounds. Performance stability was also investigated for high concentrations of CO 2 , revealing a reduction in CO 2 /CH 4 mixed-gas selectivity without compromising CO 2 permeability, suggesting strong sorption of CO 2 but minimal plasticization effects for short testing periods. In addition, time-dependent plasticization shows negligible effects on CO 2 /CH 4 mixed-gas selectivity despite an increase in CO 2 permeability when exposed to high concentrations of plasticizing CO 2 over an extended period of 170 h. This study provides valuable insights into hyperaged CANAL polymers and their performance in practical industrial processes.

Ultra-selective CANAL Polymers for Hydrogen-based Membrane Separations after Long-Term Aging

Hydrogen is a critically important molecule in the chemicals and energy industries, driving the need for efficient purification technologies such as membrane-based separations. However, polymer membranes often suffer from physical aging, leading to decreased gas permeability over time. This study leverages the unique aging behavior of contorted microporous polymers, synthesized via catalytic arene-norbornene annulation (CANAL) polymerization, to enhance molecular sieving for H 2 separations. While prior studies demonstrated a 1000% increase in H 2 /CH 4 selectivity with minimal permeability loss over 150 days, we extend this investigation to hyperaged (∼1 year) CANAL polymers and evaluate their performance under various conditions of industrial relevance. We report a remarkable 6100% increase in H 2 /CH 4 selectivity and a 2400% improvement in H 2 /N 2 selectivity, with only a 55% decline in H 2 permeability. These membranes exhibit excellent long-term stability in binary and ternary gas mixtures and at elevated temperatures. Additionally, extended aging enhances H 2 /CO 2 selectivity beyond the 2008 upper bound due to improved size-sieving. High-temperature permeation experiments and grand canonical Monte Carlo simulations reveal that H 2 permeability increases from 570 to 3500 barrer as the temperature rises from 35 °C to 190 °C, while H 2 /CO 2 selectivity declines from 14 to 3 due to the higher diffusion activation energy of CO 2 . Despite this tradeoff, hyperaged CANAL membranes maintain performance beyond the 200 °C upper bound, having selectivity–permeability performance comparable to many state-of-the-art membranes for H 2 /CO 2 separations. As a result, this study highlights the robustness and industrial viability of ultra-selective CANAL polymers for hydrogen purification, contributing to advancements in clean energy and sustainable separation technologies.

CANA v1.0.0: efficient quantification of canalization in automata networks

The biomolecular networks underpinning cell function exhibit canalization, or the buffering of fluctuations required to function in a noisy environment. We present a new major release of $\tt{CANA}$, v1.0.0, an open-source Python package for understanding canalization in automata network models, discrete dynamical systems in which activation of biomolecular entities (e.g. transcription of genes) is modeled as the activity of coupled automata. One understudied putative mechanism for canalization is the functional equivalence of biomolecular regulators (e.g. among the transcription factors for a gene). We study this mechanism using the theory of symmetry in discrete functions. We present a new exact method, $\tt{schematodes}$, for finding maximal symmetry groups among the inputs to discrete functions, and integrate it into $\tt{CANA}$. The $\tt{schematodes}$ method substantially outperforms the inexact method of previous $\tt{CANA}$ versions both in speed and accuracy. We apply $\tt{CANA}$ v1.0.0 to study symmetry in 74 experimentally supported automata network models from the Cell Collective (CC) repository. The symmetry distribution is significantly different in the CC than in random automata with the same in-degree (connectivity) and bias (average output) (Kolmogorov–Smirnov test, P ≪ .001). Its spread is much wider than in a null model (IQR 0.31 versus IQR 0.20 with equal medians), demonstrating that the CC is enriched in functions with extreme symmetry or asymmetry.

Uptake and elimination of 137Cs in aquatic biota inhabiting a contaminated effluent canal

Radiocesium (137Cs) is among the most prevalent and concerning radionuclide released from anthropogenic activities due to its relatively slow decay time and high bioavailability. Numerous studies have reported activity concentrations of 137Cs in exposed biota, but few have assessed detailed toxicokinetics. The objective of our study was to quantify and compare the uptake and elimination of 137Cs in aquatic taxa from multiple classes with different ecological roles inhabiting a canal contaminated from nuclear production activities on the Savannah River Site in South Carolina, USA. We tracked accumulation and elimination of 137Cs in four understudied aquatic species: one aquatic macrophyte [American white water lily (Nymphae odorata), two benthic associated omnivorous species (American bullfrog (Lithobates catesbeianus) and red swamp crayfish (Procambarus clarkii), and one pelagic carnivorous fish (Eastern mosquitofish (Gambusia holbrooki)]. We monitored accumulation of 137Cs from initial exposure until equilibrium for biota restricted within the canal and subsequently tracked elimination of 137Cs over time by moving organisms from the canal to a nearby reference pond. Results indicated bullfrog tadpoles reached 95 % equilibrium in approximately 9 days, the fastest among the species evaluated. After bullfrog tadpoles, the next fastest time to equilibrium was achieved by red swamp crayfish (51 days) followed by mosquitofish (86 days). At equilibrium, bullfrog tadpoles were found to have similar 137Cs activity concentrations to mosquitofish (3.16 and 3.57 Bq/g, respectively), and both were significantly higher than red swamp crayfish (1.77 Bq/g). For elimination, we found biological half-lives to range between 8 and 43 days with bullfrog tadpoles eliminating 137Cs the fastest followed by water lilies, mosquitofish, and red swamp crayfish. Overall, we found the uptake and elimination of 137Cs to vary widely among our studied species, but benthic organisms are not at an increased risk to accumulate 137Cs compared to organisms in less frequent contact with the sediment.

Pivotable support structure for cross canal solar array

A cross canal support structure for photovoltaic cells is disclosed. The structure includes a major frame having disconnectable and hinged connections to anchors at its corners, on either side of the canal. The major frame carries a plurality of minor frames, which are inclinable at an angle with respect to the major frame with the use of fixed or adjustable mounting plates. The combination of the major frame's tilt and the minor frame tilt enables fabrication of support structures that hold panels at latitude inclination for various portions of a canal.

Improvements to In-Canal Assay of High Specific Activity 60Co at the ATR

Improvements to the quality of measurements in the assaying of High Specific Activity (HSA) cobalt 60 produced at the Advanced Test Reactor at the Idaho National Laboratory are presented. Such assays are conducted regularly between cycles at the ATR, and due to the radiological safety concerns, they are performed within the ATR canal. A standard ion chamber detector was placed in a collimated assay tube within the canal. Another tube, parallel to the detector, was used to position various cobalt 60 standards and targets. The collimated port between these tubes allows for the targets to be pulled across via a winch system. Previous measurements were performed at discrete 1.5-inch steps using a hand-cranked winch system. Charge was collected and integrated for 10 seconds at each discrete step during a scan. Two substantial improvements to the assay include an electronically controlled, motor-driven winch system and a software controlled continuous charge collection. Combined, these changes improved the quality of measurements as well as streamline the assay process. Further examination of these charge profiles indicated a reasonable amount of photon scattering, or shine, before a line-of-sight is established. This created inaccuracies in the reported activity of the cobalt targets. A series of MCNP-6.3 calculations were performed in an effort to quantify and correct for this shine effect. The results of the calculations and how they were employed to a create shine correction factor for the in-canal cobalt assay at the ATR are presented. The overall bias showed an overestimation of the HSA cobalt 60 activity by around 7-8%.

Regioisomeric Spirobifluorene CANAL Ladder Polymers and Their Gas Separation Performance

For this work, we synthesized and characterized two isomeric microporous hydrocarbon ladder polymers from catalytic arene norbornene annulation (CANAL) of regioisomeric bis-norbornene fused spirobifluorenes, where the ladder chains are connected either through the same fluorene unit or across two different fluorene units in spirobifluorene. This pair of isomeric polymers was used to investigate the effect of ladder macromolecular structures on the microporosity and transport properties. Both polymers form mechanically intact films with thermal stability up to 480 °C and relatively high BET surface areas. The polymer formed from 2,7-dibromospirobifluorene showed higher BET surface area and higher gas permeability than the polymer from 2,2'-dibromospirobifluorene despite similar intersegmental spacing as indicated by X-ray scattering. The aging behavior for both polymers followed the same trend as the previously reported CANAL-fluorene polymers, with dramatically increased permselectivities over time, resulting in gas separation performance above the 2008 upper bounds for H 2 /CH 4 and O 2 /N 2 .

Canalization of Phenotypes—When the Transcriptome is Constantly but Weakly Perturbed

Abstract Recent studies have increasingly pointed to microRNAs (miRNAs) as the agent of gene regulatory network (GRN) stabilization as well as developmental canalization against constant but small environmental perturbations. To analyze mild perturbations, we construct a Dicer-1 knockdown line (dcr-1 KD) in Drosophila that modestly reduces all miRNAs by, on average, ∼20%. The defining characteristic of stabilizers is that, when their capacity is compromised, GRNs do not change their short-term behaviors. Indeed, even with such broad reductions across all miRNAs, the changes in the transcriptome are very modest during development in stable environment. By comparison, broad knockdowns of other regulatory genes (esp. transcription factors) by the same method should lead to drastic changes in the GRNs. The consequence of destabilization may thus be in long-term development as postulated by the theory of canalization. Flies with modest miRNA reductions may gradually deviate from the developmental norm, resulting in late-stage failures such as shortened longevity. In the optimal culture condition, the survival to adulthood is indeed normal in the dcr-1 KD line but, importantly, adult longevity is reduced by ∼90%. When flies are stressed by high temperature, dcr-1 KD induces lethality earlier in late pupation and, as the perturbations are shifted earlier, the affected stages are shifted correspondingly. Hence, in late stages of development with deviations piling up, GRN would be increasingly in need of stabilization. In conclusion, miRNAs appear to be a solution to weak but constant environmental perturbations.

Quantifying Nitrogen Bioextraction by Seaweed Farms – A Real-time Modeling-Monitoring Case Study in Hood Canal, WA

Anthropogenic nitrogen input is the leading cause of increasing marine eutrophication. Cultivating seaweeds at large scales may provide a viable solution for mitigating eutrophication especially when nitrogen extraction is achieved as an added environmental benefit of biofuel and food production. As the first step to promoting and implementing this solution, it is critical to quantify and assess the effectiveness of nitrogen removal by existing seaweed farms. Pacific Northwest National Laboratory and Hood Canal Mariculture Inc. conducted a case study by applying a coupled modeling-monitoring approach to quantify nitrogen bioextraction by a one-hectare commercial sugar kelp farm in Hood Canal, WA. Through this two-year effort, the project team conducted intensive field monitoring of water quality parameters at the farm site and subsequent laboratory analysis. Meanwhile, the team applied a coupled hydrodynamic-macroalgal growth model to simulate kelp growth and nitrogen bioextraction over the two kelp growing seasons. The model results showed an overall good comparison with field monitoring data and showed the effectiveness of nitrogen removal by kelp farms. The established modeling-monitoring approach can also be transferable for supporting broader research activities in macroalgal farming and eutrophication mitigation.

Control System Upgrade for Canal ARC SAW

Upgrading the specific components of the Arc Saw, such as the HMI, the components of the chassis involving the I/O modules, servo motor modules, and PLC modules offers numerous advantages. By enhancing the HMI, the operators will benefit having a user-friendly interface, enabling smoother control and monitoring of the ARC SAW’s functionality. Upgrading the modules will contribute to an improved precision and responsiveness in changing modes of the ARC SAW as well as the adjusting the saw blades position, further enhancing the system's overall performance. These upgrades, while maintaining the same functionality, enable the latest technological advancements without needing significant changes or disruptions to existing operations.

Using a Customized Portable Deepwater Portable Electrofisher to Assess Larval Lamprey Populations in Irrigation Canals

Larval lamprey densities were quantified at two large-scale water diversions facilities located on the Yakima River near the city of Yakima, Washington. Water diversions are very common in the Yakima River Basin and provide water to irrigate a diverse range of valuable cropland. Out-migrating larval Pacific Lamprey are vulnerable to entrainment through fish protection screens during the withdrawal period and desiccation or predation during dewatering at the end of the season. In order to determine the impact to lamprey populations which inhabit these regions a portable deepwater electroshocking system was utilized to determine lamprey densities which could otherwise not be determined due to water depth. Surveys were conducted in the fall during the dewatering period in 2015 and 2017 and larval lamprey densities ranged from 0.8 to 8.8 fish m-2 at the Sunnyside Diversion Facility to 4.2 to 6.5 fish m-2 at the Wapato Diversion Facility. The estimates for the total lamprey numbers at each of the region using a portable deepwater electrofishing platform (PDEP) was very similar to those estimates using backpack electrofishing post-dewatering; the PDEP method was 12- 36% higher where comparisons were available. During 2017, the estimated lamprey entrainment at the Sunnyside Diversion increased by 484% from August to November. Lamprey lengths were also determined and ranged from 50 to 130 mm. Our results indicate that the use of the deepwater shocking system was very safe and effective at determining larval lamprey densities at hard to sample regions which are present near irrigation facilities.

Enhanced Resistance Pines for Improved Renewable Biofuel and Chemical Production (Technical Report)

We completed phenotyping constitutive and inducible oleoresin flow across two seasons, constitutive resin canal number and density and wood terpene content in our ADEPT2 and CCLONES populations. We completed genetic association between 19 oleoresin phenotypes and a total of 523,192 SNP markers from ADEPT2 and 13,883 SNP markers in CCLONES using four mixed linear models. A total of 293 significant SNPs (FDR = 0.20) were identified. We used the MENTOR tool to mine mechanistic connections from a multiplex network constructed from poplar multi-omic data to construct a conceptual model for a subset of these significant SNPs. Our model contains 6 transcriptional regulators in addition to 3 monoterpene synthases. To generate more lines of evidence for these significant SNPs, we completed a time course RNAseq experiment after inducing vascular zone cells to differentiate into new resin canals with a methyl jasmonate treatment, a single nuclei RNAseq that identified differentiating resin canal epithelial cells and are completing analysis for a QTL study in a hybrid pine population. The time course identified 4634 significantly down and 1890 significantly up regulated transcripts after treatment with methyl jasmonate, an inducer of new resin canal formation in the vascular cambial meristem. To analyze this large set of differentially regulated genes, we created a predictive expression network and analyzed it with random walk restart using 6 seed genes coding for transcription factors regulating xylem differentiation in poplar. Of the top ranked 200 transcripts, 119 transcripts were significant differentially expressed supporting these transcripts as potential candidates regulating resin canal formation. Analysis of single nuclei sequencing of shoot tips that contain differentiating resin canals, identified 10 clusters. One cluster was highly enriched in transcripts coding for 9 of the enzymes in the MEP pathway 3 prenyl synthetases, and 3 monoterpene synthases strongly suggesting that this cluster represents resin canal epithelial cells. We are mining the additional transcripts to create a trajectory analysis. In summary, we have identified > 10 novel genes that are strongly supported candidates for further analysis in breeding lines and for genetic engineering over- and under- expressing lines to increase wood terpene content to improve resistance to insect and fungal pathogens while simultaneously increasing terpene supplies for renewable chemicals and biofuels.
