Draft Impacts: Modeling and Lab Validation (CRADA Final Report)

Chimney draft is a significant source of variability in real-world wood heater performance, yet laboratory tests conducted for certification do not capture this variability. Because draft varies with climate, chimney height, and home conditions, a heater that performs well in the lab can perform quite differently when installed in a home. Until now, wood heater manufacturers and installers have lacked the tools needed to anticipate how draft will vary across real installations or to advise on corrective measures when draft is too low or too high. The goal of this project was to develop and validate an open-source draft prediction tool for cordwood heater chimney systems. Over 18 months, Lawrence Berkeley National Laboratory (LBNL) and the Hearth, Patio & Barbecue Association (HPBA) collaborated to build a physics-based, Python tool that predicts how chimney draft evolves during operation, from ignition through steady burning. HPBA convened a stakeholder group of manufacturers and venting experts who provided feedback throughout development. LBNL validated the tool against laboratory measurements from a catalytic and a non-catalytic cordwood heater operated across a range of chimney heights and room-pressure conditions.

Draft Feasibility Assessment for Use of AI in Preparing Transportation Safety Analysis Reports

Preparing transportation safety analysis reports for microreactors is time and labor intensive, requiring extensive cross referencing to Federal regulations, previously approved documents, and expert review comments across structural, thermal, criticality, shielding, containment, and security. These burdens are magnified by the novelty of microreactor technologies and the evolving regulatory landscape, as well as current workforce constraints. Generative AI and supporting machine learning tools present an opportunity to accelerate drafting timelines, lift generalized writing burdens, and systematically enforce regulatory adherence through retrieval augmented generation and other knowledge retrieval and mapping methods. This draft report presents a preliminary feasibility assessment of the use of AI to expedite the preparation of microreactor transportation safety analysis reports and proposes an initial methodology for doing so.

Draft ASME Code Case to qualify L-PBF 316H material for Section III, Division 5 applications

This report documents the AMMT program’s development and submission of a draft ASME Code Case to qualify Laser Powder Bed Fusion (L PBF) Type 316H stainless steel for Section III, Divi-sion 5 Class A and SM high temperature nuclear applications. It summarizes the technical basis, the comprehensive high temperature mechanical test database assembled between 2023–2026, and the proposed code language and qualification framework submitted to ASME. The work was co-ordinated across multiple national laboratories and leverages prior ASME efforts to integrate additive manufacturing into the Boiler & Pressure Vessel Code. The body of the report describes the experimental database and analysis supporting the Code Case: tensile, creep, fatigue, creep fatigue, and thermal aging tests collected from multiple additive manufacturing sites, machine types, and powder lots, with material processed by a solution anneal heat treatment. The dataset — including both full size and subsized specimens and tests oriented parallel and perpendicular to build direction — shows limited tensile anisotropy, tensile properties comparable to wrought 316H, creep strength within the scatter of wrought material, but markedly reduced creep ductility above about 650 °C associated with rapid σ phase formation in L PBF microstructures. The draft Code Case itself prescribes a staged qualification model (manufacturing process qualification, component qualification, and per build witness testing), treats L PBF components as equivalent to Type 316 weld metal for design and inspection, and requires mechanical, chemical, and metallographic controls tied to ASTM/ISO 52946. Key acceptance criteria include tensile tests within a 90% prediction interval of the AMMT dataset, a creep fatigue screening test adapted from ASME Section III, Division 5, Subsection HB, HBB 2800 but with the cycle acceptance reduced to 100 for L PBF material, and double volumetric inspection of production components. The report concludes that the present data support treating L PBF 316H as analogous to conventional fusion weld metal for Division 5 design and inspection, while highlighting important caveats: the σ phase driven loss of creep ductility above ~650 °C, preliminary indications of enhanced creep fatigue sensitivity in some lots, and remaining gaps in long term aging and additional cyclic testing. Recommended next actions include completing outstanding cyclic and long duration creep/aging tests on the solution annealed condition, supporting inclusion of the 316H chemistry and heat treatment in ASTM/ISO 52946, and continuing engagement with ASME and NRC during balloting and review to enable industry adoption.

Draft Genome Sequences of 14 Bacterial Isolates from the Rhizosphere of Bioenergy Sorghum

We report the draft genomes of a collection of 15 bacteria, isolated from the rhizosphere soil of bioenergy sorghum (Sorghum bicolor (L.) Moench). These isolates belong to the genera, Acidovorax, Nocardioides, Agrobacterium, Peribacillus, Caulobacter, Cupriavidus, Pseudomonas, Rhizobium, Sphingomonas, Priestia, Dyadobacter, Roseomonas, Ideonella, and Bacillus.

Draft genome sequence of Gleimia europaea DSM 26657

Here, we report the draft genome sequence of Gleimia europaea DSM 26657, a pathogenic gram-positive bacillus, isolated from a patient with a subcutaneous fistula in 2007 in Germany. The genome is 2.0 Mb in size with 1,813 predicted genes, having only one putative antibiotic resistance gene.

Draft assemblies for three geographically distinct isolates of the human-infecting microsporidium Encephalitozoon intestinalis

Encephalitozoon intestinalis is an obligate intracellular parasite that causes enteritis, bronchitis, conjunctivitis, and/or encephalitis in immunosuppressed and immunocompetent individuals. To better assess its genetic diversity, here we report near-chromosome-level draft genome assemblies for three geographically distinct isolates, quadrupling the number of genome assemblies available for the enigmatic fungal pathogen.

IROS 2023 Workshop Report: Draft Guidelines on Manufacturing Procedures, Test Methods and Reporting for Soft Robotics

Soft roboticists are facing challenges with reproducibility, which prevents researchers from making holistic comparisons to prior work, impedes full understanding of results, and forces the need to “reinvent the wheel,” delaying fundamental advances. Reproducibility of results is key to advancing science as well as achieving technology transfer from research laboratories to industrial applications. Recently, a discussion-based workshop dedicated to the topic, “Developing Standard Testing and Reporting Guidelines for Soft Robotics,” was held at IROS 2023 in Detroit, MI. The purpose of this document is to record a set of recommendations and voluntary draft guidelines for soft roboticists concerning fabrication/manufacturing, test procedures, and reporting, which were collectively developed at the workshop. Together as a community, we hope to improve the reporting standards of soft robotics and drive the field as a whole toward more rigorous research practices.

Human RNome Project draft human RNome sequence of GM12878, B-cell line, obtained by mass-spectrometry sequencing, long-read sequencing and short-read sequencing.

Here we report the first draft of the human RNome sequence, a reference map of RNA chemical modifications in a human B-cell line. RNA carries a diverse repertoire of chemical modifications that regulate gene expression, cellular function, and responses to physiological and pathological cues. Yet, unlike the genome, no reference map of RNA modifications is available for any human cell. To generate this resource, the Human RNome Project Consortium analyzed a shared RNA preparation from the well-characterized GM12878 B-cell line using short-read sequencing, long-read direct RNA sequencing, and mass spectrometry, generating more than 7.1 billion sequencing reads spanning approximately 1.2 trillion nucleotides. The resulting maps of the human RNome reveal that RNA modifications are organized according to function, transcript architecture, and cellular identity. Modifications concentrate at functional centers of ribosomal and transfer RNAs, follow the canonical topology of N6-methyladenosine in coding transcripts, and form coordinated hotspots in immune regulatory genes. This first reference human RNome provides a foundation for understanding how RNA chemistry shapes cellular identity, human disease, and the development of RNA-based therapeutics.

KBase Narrative - Draft Genome of Roseomonas Strain GC11

This narrative was used for the draft genome of a Roseomonas species Strain GC11. This genus includes 6 known species. The specimen was collected using a water grab method in Conservancy Park, a freshwater site in Belleview, KY. This species is often identified in aquatic environments and is a known pathogen to humans1. It has been reported in healthcare settings from bacteremia in the blood and has also been reported to be found in human catheters. Due to the the prevelance as a human pathogen and lack of isolation in a non-human environment this strain was of particular interest.

High-quality draft genome sequence of Thermobifida halotolerans DSM 44931

Here, we report the genome sequence of Thermobifida halotolerans DSM 44931, a bacterium that was originally isolated from a salt mine in the Yunnan Province of China. This genome was sequenced using Pacific Biosciences sequencing technology and was assembled into 2 contigs in 2 scaffolds. It has a total length of 5,506,851 bp and a GC content of 71.16%. Functional annotation of this genome provides further metabolic insight into this species.
