The engineering of microbes for biomanufacturing (e.g. of fuels, chemicals, materials) applications has advanced to a stage where researchers screen genetic libraries with millions of variations each for those with enhanced productivity. This screening, however, can be slow and expensive, as screening individual variants in a high-throughput yet cost-effective manner is challenging. In this project, we aimed to reduce by 3-fold costs associated with the sequencing aspects of the screening process (to determine which genetic variant is responsible for an observed change in productivity), while being able to process over 1,000 samples per batch.

Irradiation of linear polyethylene - Partitioning between sol and gel.

Molecular weight fractions of linear polyethylene were irradiated at 133 C, in the completely molten and highly crystalline states, for the purpose of assessing the importance of chain-scission processes and establishing the critical conditions for gelation. The partitioning between sol and gel in either state was found to adhere to the theory for the intermolecular cross-linking of monodisperse species for dosages just beyond the gel point. Deviations from theory occurred as the dosage was increased further. It was concluded that main-chain scission, at these temperatures, is not a significant process. High molecular weight samples in the completely molten state obeyed the Flory-Stockmayer condition for critical gelation.

Elemental abundances in the Havero meteorite.

Abundances of 15 major, minor, and trace elements have been determined in powders and interior chips derived from the Havero ureilite. The values are in close agreement with mean values for other ureilites, as reported in the literature. The powdered samples exhibited slightly higher abundances of O and Si than the interior chips. It is suggested that this may be due to alteration of the samples during the powdering process.

A new approach to mass spectrometer measurements of thermospheric density

The gas sampling problem in satellite and high velocity probes was investigated by applying the theory of a drifting Maxwellian gas. A lens system using a free stream ion source was developed and experimentally evaluated over the pressure range of 0.00001 to 0.01 N/m sq (approx. 10 to the minus 7th power to 0.0001 torr). The source has high beam transparency, which minimizes gas-surface collisions within, or near, the ionization volume. It is shown that for high ion energy (60 eV), the extracted ion beam has an on-axis energy spread of less than 4 eV, and that 90 percent of the ions are within 2.5 deg of the beam axis. It is concluded that the molecular beam mass spectrometer concept, developed for gas density measurements in the upper atmosphere, substantially reduces gas-surface scattering and gas-surface reactions in the sample, and preserves the integrity of the gas sample during the analysis process. Studies show that both the Scout and Delta launch vehicles have adequate volume, control, velocity, and data acquisition capability necessary to obtain thermospheric number density in real time.

SEASAT economic assessment

The SEASAT program will provide scientific and economic benefits from global remote sensing of the ocean's dynamic and physical characteristics. The program as presently envisioned consists of: (1) SEASAT A; (2) SEASAT B; and (3) Operational SEASAT. This economic assessment was to identify, rationalize, quantify and validate the economic benefits evolving from SEASAT. These benefits will arise from improvements in the operating efficiency of systems that interface with the ocean. SEASAT data will be combined with data from other ocean and atmospheric sampling systems and then processed through analytical models of the interaction between oceans and atmosphere to yield accurate global measurements and global long range forecasts of ocean conditions and weather.

Evaluation of NASA speech encoder

Techniques developed by NASA for spaceflight instrumentation were used in the design of a quantizer for speech-decoding. Computer simulation of the actions of the quantizer was tested with synthesized and real speech signals. Results were evaluated by a phometician. Topics discussed include the relationship between the number of quantizer levels and the required sampling rate; reconstruction of signals; digital filtering; speech recording, sampling, and storage, and processing results.

Mixing of two liquid metals on spar payload due to spin-up and spin-down

Unexpected results from SPAR experiments, directed at obtaining fine dispersions of metal systems that in a 1-g environment tend to segregate very rapidly, were considered. The physical processes occurring in fluid samples, such as the SPAR samples before solidification, were shown to be insufficient to produce a well mixed liquid by the time solidification was initiated. This would result in solidified samples with the type of segregation noted in the SPAR samples. Experimental evidence and analytical arguments are presented.

The Viking seismometer

The Viking seismometer, operating on the Martian surface in the region of Utopia Planitia, includes three inertial velocity transducers, amplifiers, filters, automatic event detectors, data compactors, and temporary data storage capability within a unit weighing 2.2 kg. The seismometer functions at a peak magnification of 218,000 with a ground resolution of 2 nanosec and 3 Hz. Since the telemetry capacities associated with the system are limited, the seismometer instrument package employs three data processing modes: a high sampling rate mode; a heavily filtered background monitoring mode; and a frequently-used data compacting mode.

A comparative study of Conroy and Monte Carlo methods applied to multiple quadratures and multiple scattering

An efficient numerical method of multiple quadratures, the Conroy method, is applied to the problem of computing multiple scattering contributions in the radiative transfer through realistic planetary atmospheres. A brief error analysis of the method is given and comparisons are drawn with the more familiar Monte Carlo method. Both methods are stochastic problem-solving models of a physical or mathematical process and utilize the sampling scheme for points distributed over a definite region. In the Monte Carlo scheme the sample points are distributed randomly over the integration region. In the Conroy method, the sample points are distributed systematically, such that the point distribution forms a unique, closed, symmetrical pattern which effectively fills the region of the multidimensional integration. The methods are illustrated by two simple examples: one, of multidimensional integration involving two independent variables, and the other, of computing the second order scattering contribution to the sky radiance.

Transistor step stress program for JANTX2N2920

Data on the effect of power/temperature step stress when applied to the dual transistor JANTX2N2920 manufactured by Fairchild and National Semiconductor are presented. Fifty one (51) samples from Fairchild and 44 from National Semiconductor were submitted to the process outlined. In addition, two control sample units were maintained for verification of the electrical parametric testing.

The definition of hydrologic model parameters using remote sensing techniques

The reported investigation is concerned with the use of Landsat remote sensing to define input parameters for an array of hydrologic models which are used to synthesize streamflow and water quality parameters in the planning or management process. The ground truth sampling and problems involved in translating the remotely sensed information into hydrologic model parameters are discussed. Questions related to the modification of existing models for compatibility with remote sensing capabilities are also examined. It is shown that the input parameters of many models are presently overdefined in terms of the sensitivity and accuracy of the model. When this overdefinition is recognized many of the models currently considered to be incompatible with remote sensing capabilities can be modified to make possible use with sensors having rather low resolutions.

Production and analysis of thermal decomposition products from polymeric materials

A description is presented of a strategy for analyzing the combustion process and the degradation products which are formed. One of three primary objectives in the study of polymer degradation is related to the characterization of the material to be studied and the investigation of the thermal behavior of the material. Another objective is concerned with the definition of the nature of the decomposition process by identification and quantitation of the degradation products. The third objective involves the determination of the mechanism and kinetics of the decomposition process. The methods of sample degradation include pyrolysis, oxidative degradation, flaming combustion, and the use of large-scale combustion chambers. Methods of chemical separation and identification are considered, taking into account low-boiling volatiles, high-boiling volatiles, and ancillary techniques.

Airborne laser induced fluorescence system for measuring OH and other trace gases in the parts-per-quadrillion to parts-per-trillion range

Described in detail is a laser induced fluorescence system which has been successfully interfaced with two aircraft sampling platforms (i.e., Sabreliner jet and an L-188C Electra). This system, which has been under development for four years, presently consists of the following major components: (1) a Nd-Yag laser driven oscillator-amplifier dye laser; (2) a sampling manifold with associated fluorescence detection optics; (3) an OH calibration chamber; (4) a laser beam steering assembly; and (5) sampling electronics and data processing hardware. During the last three years, this system has been flown some 50,000 air miles making tropospheric OH radical measurements over the latitude range of 70 N to 57 S. OH concentrations measured during these flights have ranged from 30 parts-per-quadrillion (3.7 x 10 to the 5th molecules/sq cm) at altitudes of 6 km to 0.8 parts-per-trillion (2.0 x 10 to the 7th molecules/sq cm) at 0.5 km. Computations have been completed which indicate that the existing aircraft system with modest modifications should also be capable of detecting natural tropospheric levels of NO, SO2, CH2O, NO2, HNO2, NO3, H2O2 and CS2 by using both conventional laser-induced fluorescence methodology and multiphoton techniques.
